Objective: To observe the effects of apigenin(API) on the growth and apoptosis of cervical cancer HeLa cells and to investigate the mechanism underlying whether apigenin inhibits growth and induces apoptosis of cervical cancer HeLa cells is involved in inhibition of CK2α protein expression and blocking Notch1 Signaling pathway.Methods: The plate colony formation method was performed to detect the cell colony forming ability of cervical cancer HeLa cells treated with different concentration of apigenin for different time. To analysis the apoptosis rate of cervical cancer HeLa cells treated with different concentration of apigenin for 24 h, flow cytometry using Annexin-V/PI double staining was conducted. The expressions of CK2α, Notch1 and Hes1 of cervical cancer HeLa cells treated with different concentration of apigenin for 24 h were analyzed by Western blot.Results: 1. The results of plate colony formation assay showed that different concentration of apigenin(10.0, 20.0, 40.0, 80.0 mg/L) inhibited the growth of cervical cancer HeLa cells significantly, the colony inhibition rate increased gradually of cervical carcinoma HeLa cells treated by apigenin at different time(12 h, 24 h, 48 h), in time- and dose- dependent manner, comparison of experimental drug treatment group and the control group, the differences were statistically significant(P < 0.05). 2. Data of flow cytometry analysis with Annexin-V/PI doubl e staining revealed that untreated group(blank control group), 0.1%DMSO group(vehicle control group), apigenin(10.0 mg/L) group, a pigenin(20.0 mg/L) group, apigenin(40.0 mg/L) group and apigenin(80.0 mg/L) group for 24 hours, the apoptosis rate of cervical can cer HeLa cells were 1.57±0.06%, 1.67±0.07%, 3.77±0.06%, 5.57±0.21%, 11.00±0.20% and 16.87±0.15%, respectively. Experimental drug treatment group compared with the control group, the difference w as statistically significant(P<0.05). 3. Western blot showed that apigenin can down-regulate the expression of CK2α, Notch1 and Hes1 of cervical cancer HeLa cells in a dose-dependent manner(P < 0.05).Conclusion: Apigenin can inhibit the growth and induce apoptosis of human cervical cancer HeLa cells and its mechanism may is involved in inhibition of CK2α protein expression and blocking Notch1 Signaling.
