| Objective:By using the quantitative analysis method of lipidomics to analyze the difference of lipid metaboli tes in common syndromes of polycyst ic ovarian syndrome(PCOS),to explore the potential biomarkers of PCOS liver qi depression syndrome The pathogenesis of liver depression and kidney deficiency syndrome.Method:Patients with clinical PCOS were divided into groups of liver stagnation and kidney deficiency syndrome,liver stagnation syndrome,kidney deficiency syndrome syndrome and serum samples were collected,and a heal t hy control group was set up.Based on ultra-high performance liquid chromatography-quadrupoletime-of-flight mass spectrometry(UHPLC-QTOF-MS),the samples were used for quantitative detection of lipid molecules,XCMS software was used for data processing and lipid composition identification.After principal component analysis,orthogonal part ial least squares-discriminant analysis,and t test for statistical analysis of the data,lipids with significant differences between groups were screened.Compare the differences of serum lipid metabolites between PCOS liver stagnation and kidney deficiency syndrome and liver stagnation syndrome,kidney deficiency syndrome and normal group,determine the pot ential biomarkers of PCOS liver stagnation and kidney deficiency syndrome,and explore the corresponding mechanism.Result:1.There are significant differences in serum lipid metabolites between PCOS liver stagnation and kidney deficiency group and normal group.Compared with the normal group,the content of PI,OxPI,OxPC,PEtOH,etc.in the PCOS liver stagnation and kidney deficiency syndrome group was lower than that in the normal group,and the lipid content of Cer-NS,Cer-AS,Cer-AP,LPC,etc.was higher than that in the normal group.Substances include fatty acids,glycerides,glycerophospholipids,sphingomyelins,and polyethylene 5 lipids.2.PCOS liver depression syndrome is relatively normal group,there are 139 lipid molecules with PI,LPI,OxPI,TAG,SHexCer,SM and other lipids have significant differences.Compared with the normal group of PCOS kidney deficiency syndrome,there are differences in 206 lipid molecules of Cer,SM,DAG,LPA,FAHFA and other lipid species.3.PCOS liver stagnation and kidney deficiency syndromes include ACar,DAG,DGTS,PC and other 82 lipid substances.There are differences between PCOS kidney stagnation syndrome and PCOS kidney deficiency syndrome;There are differences between 70 lipid substances of 25 lipid types and PCOS liver stagnation syndrome.4.PCOS liver stagnation and kidney deficiency syndrome were compared with liver stagnation syndrome,kidney deficiency syndrome,normal group,ACar(13:1),SM(d16:0/22:0),DGTS(2:0-20:3),Cer-NDS(d18:0/22:0),Cer-AP(t24:0/17:0),Cer-AP(t 14:0/25:0),are present in the three comparisons,which is the syndrome of liver depression and kidney deficiency in PCOS Unique difference substance.Conclusion:1.ACar(13:1),SM(d16:0/22:0),DGTS(2:0-20:3),Cer-NDS(d18:0/22:0),Cer-AP(t24:0/17:0),Cer-AP(t14:0/25:0)may be a potential biomarker for PCOS liver deficiency and kidney deficiency syndrome.2.PCOS liver and kidney deficiency syndrome has lipid metabolism disorder,mainly manifested as abnormal metabolism of glycerol phospholipids,glycerides and sphingomyelin.3.The internal biological mechanism of PCOS liver depression and kidney deficiency syndrome may be related to the PI3K/Akt signal transduction,liver lipid peroxidation,and vascular inflammation.Lipidomics analysis of serum can comprehensively reflect the syndrome characteristics and pathological mechanism of PCOS liver stagnation and kidney deficiency syndrome. |
