ObjectiveTo investigate the protein expression of mTOR and survivin in human cervical squamous carcinoma and analyze the relationship between the proteins and clinic pathological characters and their correlation with HPV16/18 infection of cervical squamous carcinomaMethods60 cases of cervical invasive cervical squamous carcinoma,30 cases of low-grade cervical squamous intraepithelial lesion(LSIL),30 cases of high-grade cervical squamous intraepithelial lesion(HSIL)and 30 cases of total hysterectomy due to hysteromyoma were collected from Jinzhou Women and Children’s hospital.All tumor cases were not treated with chemotherapy or radiotherapy before resection.Immunohistochemistry and Western blot were used to detect the expression of mTOR and survivin.Cervical exfoliated cells were collected for the detection of high-risk HPV16/18.The data were analyzed by SPSS 25.0.Results1.The positive rates of mTOR was higher in cervical squamous carcinoma 68.3%(41/60)than that in HSIL 46.7%(14/30),LSIL 20%(6/30)and normal cervical tissues 3.3%(1/30)(P<0.01).mTOR protein expression is related to the stage and lymph node metastasis of cervical squamous carcinoma(P<0.01).2.The positive rates of survivin was higher in cervical squamous carcinoma(76.7%,46/60)than that in HSIL 53.3%(16/30),23.3%(7/30)and normal cervical tissues(0%,0/30)(P<0.01).The survivin protein expression is related to the stage and lymph node metastasis of cervical squamous carcinoma(P<0.01).3.There was significant positive correlation between mTOR and survivin in cervical squamous carcinoma(r=0.472,P<0.01).4.The positive rates of HPV16/18 was higher in cervical squamous carcinoma(81.7%,49/60)than that in HSIL(56.7%,17/30),LSIL(26.7%,8/30)and normal cervical tissues(3.3%,1/30)respectively(P<0.05).5.The expression of mTOR and survivin protein was positively correlated with HPV16/18 infection(r=0.326,r=0.350,P<0.01),and OR=5.396,or=6.150,respectively.6.Western blot showed that the expression of mTOR and survivin protein increased gradually in the normal cervix tissues,LSIL,HSIL and cervical squamous carcinoma(P<0.01),which was consistent with the results of immunohistochemistry.Conclusions1.The protein expression of mTOR and survivin in cervical squamous carcinoma was significantly higher than that in SIL and normal cervical cancer,and was related to tumor stage and lymph node metastasis.2.HPV16/18 positive infection rate of cervical squamous carcinoma was significantly higher than that of SIL and normal cervical cancer.3.mTOR,survivin protein expression and HPV16/18 infection are positively correlated,and they may interact and participate in the development of cervical squamous carcinoma.
