Objective:To investigate the expression of osteopontin(OPN) in the organization and serum and the expression of SCC-Ag in the serum in women with Uterine cervix cancer, thereby to explore the role of them and their relationship in the pathogenesis and the signs of Traditional Chinese medicine of endometriosis. To study the relations between OPN,SCC-Ag and Chemo sensitivity, and the relationship between OPN and SCC-Ag in patients of Uterine cervix cancer, and to discuss its value of diagnosis and clinical applicationMethods:The expression of OPN in cervical tissue are detected by immunohistochemistry method in 92 cases women with cervical squamous cell carcinoma,57 cases of women with Cervical intraepithelial neoplasia of change (CIN), and 20 cases of women with Normal cervical, all the cases were treated in our hospital from 2006 to 2010.The levels of osteopontin in serum of 65 cases of women with cervical squamous cell carcinoma (22 with stage Ia1~Ib1 underwent surgery, 22 with stage Ia1~Ib1 underwent neoad-juvant chemotherapy and surgery,9 with stageⅡb~Ⅳunderwent radiation therapy),10 cases of women with cervical non-squamous cell carcinoma,34 cases of women with Cervical intraepithelial neoplasia of change(CIN), and 20 cases of women with Normal cervical from the yesr 2009 were detected by enzyme-linked immunosorbent assay(ELISA),to investigate the significance of OPN levels in tissue and in serum and compared the levels with the results between the clinicopathological stages and pre-and post-treatment, and relationship between OPN and Chemo sensitivity in squamous cell carcinoma of the cervix. And serum SCC-Ag of patients with cervical squamous cell carcinoma were detected by Chemiluminescence Microparticle Immuno Assay (CMIA), and investigate the significance between the serum SCC-Ag levels and clinicopathological stages, prognosis of squamous cell carcinoma (SCC) of the cervix, and the relationship between OPN and serum SCC-Ag levels, and relationship between SCC-Ag and Chemo sensitivity in squamous cell carcinoma of the cervix.Results:1. The expression of OPN in the squamous cell carcinoma (SCC) of the cervix higher than in the Cervical intraepithelial neoplasia of change (CIN) and Normal cervical (P<0.05). There was a significant difference in levels of OPN in the serum between squamous cell carcinoma (SCC) of the cervix group and Normal cervical. There was no significant difference between the squamous cell carcinoma (SCC) of the cervix group and the CIN group, but the levels of OPN in the serum of squamous cell carcinoma (SCC) of the cervix group higher than the CIN group.2. The expression of OPN in tissue of squamous cell carcinoma (SCC) of the cervix with the FIGO stage, tumor invasive depth, lymph node metastasis and cell differentiation. The levels of OPN inⅢ-Ⅳstage higher than that inⅠ~Ⅱstage, The levels of OPN in pathological type was low-differentiated squamous cell carcinoma higher than that in pathological type was moderately or well-differentiated, The levels of OPN in tumor invasive depth over 1/2 interstitial grop higher than that less than 1/2 interstitial grop, Significantly higher levels were associated with lymph node metastases. There was no significantly difference in different age of the patients.3. SCC-Ag expression in serum associate with clinical stage of cervical squamous cell carcinoma, tumor invasive depth, lymph node metastasis. The levels of SCC-Ag inⅢ~Ⅳstage higher than that inⅠ~Ⅱstage, The levels of SCC-Ag in pathological type was low-differentiated squamous cell carcinoma higher than that in pathological type was moderately or well-differentiated, The levels of SCC-Ag in tumor invasive depth over 1/2 interstitial grop higher than that less than 1/2 interstitial grop, Significantly higher levels were associated with lymph node metastases. There was no significantly difference in different age of the patients and different tumor histological type.4. The levels of OPN and SCC-Ag the squamous cell carcinoma (SCC) of the cervix lower than before after neoad-juvant chemotherapy. There were significantly difference of SCC-Ag in serum pre-and post neoad-juvant chemotherapy in the effective group, and no significantly differdence in the uneffective group.5. The consistency of OPN and SCC-Ag expression results show that, as the disease progresses, the OPN and SCC-Ag are both higher. both expression of OPN and SCC-Ag were positively correlated in cervical squamous cell carcinoma.6. There were no statistically significant differences of OPN in tissues and in serum in different TCM syndrome differentiations, There were statistically significant differences of serum SCC-Ag in TCM syndrome differentiations which And card stomach and Spleen and kidney Yang syndrome, And card stomach and Liver-kidney Yin deficiency syndrome, Hot and humid stasis poison card and kidney Yang syndrome, Hot and humid stasis poison card and Liver-kidney Yin deficiency syndrome. There were no statistically significant differences of SCC-Ag in other different TCM syndrome differentiations.Conclusion: Overexpression of OPN SCC-Ag may play an important role in the occurrence,transformation,development and prognosis of cervical squamous cell carcinoma. Expression of OPN was positively correlated in cervical squamous cell carcinoma and significant relationship between OPN and Chemo sensitivity in squamous cell carcinoma of the cervix, and the level of OPN in serum can show the expression of OPN in tissue. It was hihgten with the disease progresses. The level of SCC-Ag in serum can show the degree of cervical squamous cell carcinoma. The level of OPN and SCC-Ag in serum associate with clinical stage of cervical squamous cell carcinoma, higher level associate with higher clinical stage. It has a potential clinical significance as a prognostic marker for patients to check both OPN and SCC-Ag. The relations between OPN,SCC-Ag and TCM syndromes were expected to provide a new diagnosis and treatment of direction.
