| ObjectiveBy means of network pharmacology Siwu decoction(Siwu decoction,SWD)to study the characteristics,construction of SWD chemical composition database,confirmation of active compounds in Siwu and the corresponding and target parts,and analyze the pathogenic genes of polycystic ovary syndrome by bioinformatics,the construction of SWD-PCOS protein interaction network,analysis of SWD key targets for the treatment of PCOS and the biological function by bioinformatics,explore the underlying mechanisms of Siwu decoction to treat PCOS,It provides theoretical support and experimental basis for clinical rational drug use.MethodThe chemical constituents of Siwei Decoction in Siwu Tang were searched through the three major Chinese medicine databases of TCMID,TCMSP,and TCM @ taiwan,and the chemical composition database of Siwu Decoction was constructed.Active compounds and potential targets were screened according to OB and DL.And use Pubchem and Uniprot to unify the names of the compounds and targets.Then use NCBI-GENE and OMIM database to detect PCOS related genes,and further establish a PCOS database.The corresponding target of SWD is applied to the corresponding disease gene interaction network to detect the target of the treatment of PCOS,and then the GO and KEGG biological information analysis is carried out through the Clue GO plug-in to study the mechanism of Siwu Tang on PCOS.Result1.Through the corresponding Chinese medicine database,475 Siwutang compounds were further obtained,and OB and DL were used to further screen out the active compounds satisfying the conditions and the corresponding target targets,respectively 44 of 247;through NCBI-GENE 262 PCOS-related human genes were detected with the OMIM database;a compound-target network,a target interaction network and a gene interaction network were established respectively.Applying the drug target action network to the disease gene interaction network can further obtain 12 targets.They may be the main targets in the treatment of PCOS with Siwu Tang.Analysis of the above targets through the GO and KEGG biological pathways revealed that they can regulate apoptosis,reactive oxygen metabolism and protein kinase B information transmission;their function is to constitute alpha particles in platelets,RNA polymerase II Transcription factor and plasma membrane raft;it also has many functions,such as cell activity,interaction with protein phosphatase and stimulation of steroid hormone receptors.Analysis of key targets using KEGG biological pathway enrichment revealed that the main biological pathways of key targets include: AGE-RAGE signaling pathway,PI3K-Akt signaling pathway,HIF-1 signaling pathway,and non-alcoholic fatty liver(NAFLD)and other biological pathways.ConcludeHere,we establish the corresponding database of Siwu Tang through network pharmacology,and then obtain the corresponding PCOS target genes based on the obtained data,and further establish the compound-target,target protein interaction and disease-related protein interaction through network pharmacology As a network,study the basic mechanism of the network and its topological parameters,and discover the main mechanism of Siwu Tang in the treatment process,use the corresponding drug target network to the corresponding PCOS gene protein interaction network system,analyze and study four The basic principles generated by Wutang in the treatment of PCOS are then verified to obtain the final results.The above methods have laid a solid foundation for clinical treatment experimental research to a certain extent,and promoted the further development of traditional Chinese medicine. |
