Part One Experience miningObjective:To summarize the law of syndrome differentiation and treatment of polycystic ovary syndrome treated by Professor Wei Zixiao,a famous Chinese gynecologist in the capital of China,and extract the core prescription by using the method of data mining.In order to better learn Professor Wei Zixiao’s experience in the treatment of polycystic ovary syndrome.Methods:Collected and sorted out the outpatient case data of Professor Wei Zixiao’s treatment of polycystic ovary syndrome,standardized and sorted out the data,and then imported it into the ancient and modern medical case cloud platform(V2.2.1).By using its data mining and analysis function,the frequency statistics,correlation analysis,cluster analysis and complex network analysis were carried out for basic information,four diagnosis data,TCM syndromes,medication,etc.According to the mining results,Professor Wei Zixiao’s experience in the treatment of polycystic ovary syndrome was summed up,and the core prescription was extracted.Results:This study included initially diagnosed cases of polycystic ovary syndrome treated by Professor Wei Zixiao from January to January.The age ranges from 14 to 42 years old,and the main age range is 20 to 39 years oldaccounting for 80.6.%of the total.Symptoms were most prominent delayed menstrual cycle(41times,56.94%),insomnia(30times,41.67%),and hypomenorrhea(23times,31.94%).The main features of tongue were tooth-marked tongue,plump tongue and dark tongue.The pulse is thread and stringy.From syndrome,phlegm and dampness syndrome,Qi stagnation and blood stasis syndrome are the main syndromes.The main methods of treatment include promoting Qi and promoting blood circulation,warming the kidney and promoting Yang and reducing phlegm and dehumidification.From the point of view of traditional Chinese medicine,the high frequency of chuanxiong,xiangfu,salvia miltiorrhiza,atractylodis,tangerine peel,pinellia,spina gleditsiae,poria,aromatica.The four Qi of medicine are mainly warm and flat.Five flavors are more common,such as pungency,bitterness and sweetness.Mainly into the liver,spleen,heart,lung,kidney,etc.According to the correlation analysis between TCM syndromes and other case data,the main clinical manifestations of phlegm and dampness are hypomenorrhea,premenstrual abdominal pain,insomnia,impatience,tooth-marked tongue,reddish or reddish tongue color,thread or deep pulse.The use of drugs to pay attention to reducie phlegm and dehumidification phlegm and dehumidification and warm the kidney and promote Yang.the use of atractylodis,tangerine peel,pinellia,poria,xiangfu,chuanxiong,cinnamon,etc.The syndrome of Qi stagnation and blood stasis is mainly manifested by delayed menstrual cycle weakness,purple and dark menstrual color,acne,lumbar acid and lumbago,fat tongue,red tongue or slightly dim red tongue,white or thin moss,and pulse string.Treatment to promote Qi and activate blood-nourishing liver-based.Multi-purpose yu jin,bupleurum,salvia miltiorrhiza,angelica,saponin and other drugs.The treatment is mainly to promote Qi and blood circulation,nourish blood and dredge liver.Most of the drugs used are yujin,bupleurum,salvia miltiorrhiza,angelica,spina gleditsiae and so on.According to the association rules of traditional Chinese medicine,chuanxiong,and xiangfu,atractylodis and chuanxiong,atractylodis and tangerine peel,pinellia and chuanxiong,pinellia and tangerine peel,pinellia and atractylodis are the main compatibility.From the cluster analysis of high-frequency drugs,five groups of drug cluster results were obtained:cinnamon,chuanxiong,xiangfu,atractylodis,tangerine peel,pinellia,poria;spina gleditsiae,lycopi;dodder,angelica,salvia miltiorrhiza,aromatic,radix bupleuri and so on.Through the analysis of the complex network of drugs,the core prescription of Professor Wei Zixiao’s treatment of polycystic ovary syndrome was extracted,which was composed of chuanxiong,xiangfu,atractylodis,pinellia,tangerine peel and poria.Conclusion:Professor Wei Zixiao believes that the basic pathogenesis of polycystic ovary syndrome is the disorders of liver,spleen and kidney function,phlegm and dampness,Qi stagnation and blood stasis blocking the body and blocking the uterus.In the process of diagnosis and treatment,starting with phlegm and dampness,the treatment methods such as resolving phlegm and dehumidifying,activating Qi and promoting blood circulation,supporting vital energy,etc.are used to make the internal Qi and blood body fluid run smoothly.Part Two Discussion on the mechanism of core prescriptionObjective:To explore the effective components,targets and functional pathways of Professor Wei Zixiao ’s treatment of polycystic ovary syndrome by using network pharmacology.To explore the mechanism of core prescription.Methods:The core prescription active ingredients and corresponding targets were obtained by searching the pharmacology database and analysis platform of TCM system.The disease target of polycystic ovary syndrome was retrieved through Gene Cards database and OMIM database,and the common target of core prescription and polycystic ovary syndrome disease target was obtained after mapping the core prescription target and polycystic ovary syndrome disease target,namely the key target.Cytoscape was used to construct the core prescription drug-component-disease-target network.Through String,protein-ininteraction analysis was carried out on the common target,and the core target was screened out.Through R language and its biological package,GO and KEGG analysis was carried out on the common targets of core prescription and polycystic ovary syndrome.Results:The core prescription(chuanxiong,xiangfu,atractylodis,pinellia,tangerine peel,Poria),contains quercetin,luteolin,wogonin,baicalein,kaempferol,etc.25 active ingredients.In terms of targets,the active ingredients of the core prescription acted in combination on 39 targets of polycystic ovary syndrome,mainly including EGFR,CASP,CCND,ESR,IL6,etc.Chuanxiong has 3 active components acting on 6 targets of the disease.Xiangfu has 10 active components acting on 37 targets of the disease.Atractylodis has 4 active components acting on 9 targets of the disease.Pinellia has 6 active components acting on 14 targets of the disease.Tangerine peel has 3 active components acting on 10 targets of the disease Poria has 3 active components acting on 1 target of the disease.It can be seen that the core prescription ACTS on polycystic ovary syndrome through multiple components and targets.Through protein interaction analysis,the core targets were EGFR,CASP3,CCND1,ESR,IL6,MYC,etc.According to the GO enrichment results of the targets,the key targets were mainly enriched in nuclear receptor activity,transcription factor activity,direct ligand regulated sequence-specific DNA binding and steroid hormone receptor activity.From the KEGG enrichment results,the key targets are mainly concentrated in the pathways of Proteoglycans in cancer,Testicancer,Prostate cancer,cystic resistance,MAPK signaling pathway and PI3K-Akt signaling pathway.Conclusion:Professor Wei Zixhao’s core prescription for the treatment of polycystic ovary syndrome has the characteristics of multi-components and multi-targets,mainly through anti-inflammatory,regulation of glucose and lipid and hormone metabolism,and improvement of endometrial receptivity.
