| Objective: Based on the establishment of bronchial asthma model of liver stagnation syndrome,metabonomics method was used to determine the differential metabolites of asthma in liver stagnation syndrome.By comparing the changes of differential metabolites,we should investigate the effect of Shu Gan Li Fei on liver stagnation syndrome.The metabolomics mechanism of bronchial asthma also illustrates the advantages of traditional Chinese medicine for prescription treatment.Methods : A model of bronchial asthma with liver stagnation syndrome was established by using the recognized egg protein sensitization method and chronic unpredictable stress.70 male Wistar rats were randomly adapted and randomly divided into normal group(group A)and bronchial asthma model group.Group B),bronchial asthma liver stagnation syndrome model group(group C),bronchial asthma liver stagnation syndrome model group +dexamethasone control group(group D),rat bronchial asthma liver stagnation syndrome model group + high dose treatment group(Group E),rat bronchial asthma liver stagnation syndrome model group + medium dose treatment group(Group F),rat bronchial asthma liver stagnation syndrome model group +low dose treatment group(Group G),10 rats in each group.During the modeling period,the behavior and morphological changes of each group were observed and the changes of body weight and sugar consumption were recorded.After stopping the modeling,groups A,B and C were given 2 ml/normal saline daily;group D was 0.1 mg/100 g dexamethasone daily.The rats in the E group,the F group and the G group were given 2.79mg/100 g,5.58mg/100 g,8.37mg/100 g Shu Gan Li Fei compound for 5 days.The urine of each group before the administration and the urine of the first,third and fifth days after the administration were collected,and the samples were analyzed by LC-MS/MS,and analyzed by PCA principal component analysis and unsupervised bias.Least Squares Discriminant Analysis OPLS-DA performs supervised data analysis,screens out differential metabolites,and changes the way of comparing differential metabolites,and explores Shu Gan Li Fei and dexamethasone intervention for liver stagnation syndrome bronchial asthma The metabolomics mechanism also illustrates the advantages of traditional Chinese medicine in Shu Gan Li Fei.Results: By analyzing the 10 differential metabolites screened,it was found that the high dose group had lower histidine,serine and hippuric acid content,creatinine,cysteine and glutamine compared with the liver stagnation syndrome group.The contents of acid,citric acid,acetoacetic acid and leucine/isoleucine increased,and the difference was statistically significant(P<0.05).The middle and low dose groups were compared with the liver stagnation syndrome group.The content of acid and serine decreased,and the content of cysteine,glutamic acid,acetoacetic acid and leucine/isoleucine increased,the difference was statistically significant(P<0.05),while the dexamethasone group only The content of histidine and serine decreased,and the content of glutamic acid increased,and the difference was statistically significant(P<0.05).It was also found that except for leucine/isoleucine,all other metabolic markers had a tendency to call back to the normal group.Conclusions: It was found that histidine,hippuric acid,asparagine,creatinine,acetoacetic acid,citric acid,serine,cysteine,glutamic acid,leucine,etc.may be metabolic markers of the liver model of asthma syndrome,and Shu Gan Li Fei can regulate the metabolites to normal levels in different degrees.The high-dose treatment group can not only affect most metabolic markers,but also the most obvious trend of callback,which is obviously superior to other dose-treated groups,indicating liver-dissolving Li Fei Recipe may be involved in the regulation of energy metabolism,amino acid metabolism,fat metabolism and other metabolic pathways in the asthma model of liver stagnation syndrome,thus affecting multiple systems of the body. |
