| ObjectiveBy observing the changes of TLR4 protein,TLR4 mRNA,LPS,inflammatory factors including TNF-α,IL-6,CRP,etc.To investigate the hypoglycemic mechanism of yam gruel on T2DM rats,and provide scientific basis for the renewed research and popularization of yam gruel.MethodsThe forty-eight SPF male Wistar rats were randomly divided into 10 normal rats and 38 model rats according to body weight,which were fed with ordinary feed and high-fat and high-sugar feed,respectively.After 6 weeks fasting for 12 hour overnight,the model group was given a 25 mg/kg intraperitoneal injection of STZ buffer.72 hours after the injection of streptozotocin,FPG was measured in the tail vein.After 2 weeks of observation,the final FPG was≥11.1 mmol/L on two different days,accompanied by symptoms such as multiple drinks,more food,more urine,weight loss,can be determined as model T2DM rats.After successful modeling,30 model rats were numbered according to the level of the last FPG value.After successful modeling,30 model rats were numbered according to the level of the last FPG value.They were divided into model group,positive control group,and yam gruel group by random number table method,10 rats in each group.The normal group and the model group were given saline 15ml/kg once daily at 9:00 am,The positive control group received metformin 15ml/kg once daily at 9:00 am,the yam gruel group was fed of yam gruel 15 ml/kg once daily at 9:00 am,continuous intervention for 6 weeks.During the intervention period,FPG was monitored once a week and weight was measured once every 2 weeks.After the intervention,the expression of TLR4 protein was detected by Western-blot,and the expression of TLR4 mRNA was detected by RT-PCR.The levels of LPS,TNF-α,CRP and IL-6 in serum of T2DM rats were detected by ELISA.Results1 Establishment of rat model of T2DMCompared with the pre-modeling rats,the body weight of the rats in the model-building group decreased,and the FPG increased.The difference was statistically significant(P<0.01).Compared with the normal group,the weight of the rats after modeling decreased and the FPG increased,the difference was statistically significant(P<0.01).At the same time,the food intake,water intake,and urine output of the model group increased significantly.2 Comparison serum levels of LPS in rats after interventionCompared with the normal group,the expression level of LPS in the model group was significantly higher(P<0.05).Compared with the model group,the LPS expression level in the yam gruel group was significantly lower(P<0.01),and there was no significant difference between the positive control group and the yam gruel group(P>0.05).3 Comparison of expression levels of TLR4 protein in rats after interventionCompared with the normal group,the expression of TLR4 protein in the model group was significantly higher(P<0.01).Compared with the model group,the expression level of TLR4 protein in the yam gruel group was significantly decreased(P<0.01),and there was no significant difference in the expression level of TLR4 protein between the positive control group and the yam gruel group(P>0.05).4 Comparison of expression of TLR4 mRNA in rats after interventionCompared with the normal group,the expression of TLR4 mRNA in the model group was significantly higher(P<0.01).Compared with the model group,the expression level of TLR4 mRNA in the yam gruel group was significantly decreased(P<0.05).and there was no significant difference in the expression level of TLR4 mRNA between the positive control group and the yam gruel group(P>0.05).5 Comparison of serum levels of TNF-α,CRP and IL-6 in rats after interventionCompared with the normal group,the expression levels of TNF-α IL-6 and CRP in the model group were significantly higher(P<0.01).Compared with the model group,the expression levels of TNF-α,IL-6,and CRP in the positive control group and yam gruel group were significantly lower(P<0.05).Compared with the positive control group,the expression levels of IL-6 and CRP were lower in the yam gruel group(P<0.01).6 Comparison of FPG in rats after interventionCompared with the normal group,the FPG in the model group was significantly higher(P<0.01).Compared with the model group,the FPG in the positive control group and yam gruel group decreased significantly(P<0.05),and the positive control group and yam gruel group had no significant difference in FPG levels(P>0.05).7 Comparison of body weight of rats after interventionCompared with the normal group,the body weight of the model group increased significantly(P<0.01).Compared with the model group,there was no significant difference in weight gain between the positive control group and the yam gruel group(P>0.05).There was no significant difference in weight gain between the positive control group and the yam gruel group(P>0.05).ConclusionAfter 6 weeks of intervention with the yam gruel,the expression of TLR4 and TLR4 mRNA in the colon of T2DM rats was decreased,and the levels of serum LPS,inflammatory factors including TNF-α,IL-6,and CRP were decreased,simultaneously improves FPG and T2DM-related symptoms in T2DM rats,suggesting that the yam gruel may inhibit the expression of TLR4 signaling pathway,improve the inflammatory response,and play a hypoglycemic effect. |
