Huang Qi Decoction Ameliorates Inflammatory Response Through Blockade Of The TLR4/NF-κB Signaling Pathway Streptozotocin-induce Diabetic Rats

Objective:The inflammatory response plays an important role in the development and progression of diabetic nephropathy(DN).Therefore,reducing the inflammatory response can delay the progression of diabetic nephropathy.In this paper,we established a model of diabetic nephropathy induced by lipopolysaccharide(LPS)to observe whether Huangqi Decoction can improve the inflammatory response in rats with diabetic nephropathy by inhibiting TLR4/NF-κB signaling pathway.Methods:The Six-week-old male clean SD rats were randomly divided into 6 groups: control group,control group + LPS group,model group,model + LPS group,Huangqi decoction intervention group,Huangqi decoction + LPS group,and animal model was peritoneal cavity.Streptozotocin(STZ)was injected and the fasting blood glucose of SD rats was measured two weeks later.The fasting blood glucose was greater than 16.6 mmol/L.After successful modeling,Huangqi Decoction was administered by intragastric administration for a total of 8 weeks.LPS was intraperitoneally injected at the 7th week and sacrificed 8 weeks later.The general indicators,renal function and pathological changes of the rats were examined.The expression of TLR4/NF-κB signaling pathway-related proteins and related proteins downstream was detected by Western blot.The expression of TLR4,PNF-κB and TNFα was detected by immunofluorescence technique.Through the above technical means to observe the effect of Huangqi Decoction on the inflammatory response in rats with diabetic nephropathy,and to explore the intervention target of Huangqi Decoction in the treatment of diabetic nephropathy.Results:The TLR4/NF-κB signaling pathway was significantly activated in diabetic nephropathy rats and LPS+ diabetic nephropathy rats.The expression levels of TLR4,phosphorylated IKKα/β,phosphorylated NF-κB and other proteins were significantly increased,and downstream inflammatory factors.The expression also increased,renal pathological indicators and fibrosis increased.Huangqi Decoction can improve the general indicators,renal function and proteinuria in rats with diabetic nephropathy and LPS+ diabetic nephropathy;it can reduce the phosphorylation of IKK and NF-κB by reducing the expression of TLR4,and achieve the inhibition of TLR4/NF-κB signal.The role of the pathway reduces the expression levels of downstream inflammatory factors such as IL6,TNF-α,and IL1β,thereby reducing inflammation,improving renal pathological parameters,and reducing renal fibrosis.Conclusion:The study showed that Huangqi Decoction can improve the inflammatory response of rats with diabetic nephropathy and LPS+ diabetic nephropathy,and down-regulate the expression level of TLR4/NF-κB signaling pathway-related protein,thereby alleviating renal damage in rats with diabetic nephropathy and delaying diabetes.The role of kidney disease progression.
