Clinical Research On Effect Of Bushen Huoxue Formula On IL-1β、IL-33 Of Peripheral Blood In URSA

Objective:Recurrent abortion is a common gynecological disease,which affecting the lives of patients and their families,to bring them psychological and physical pain.Modern medicine on the recurrence of unexplained recurrent abortion,there is no certainty of norms,and the motherland medicine tocolysis in the method of Bushenhuoxue in the treatment of the disease which have good efficacy and development prospects.Chinese medicine Bushen Huoxue side is the instructor after years of clinical application summed up,has made significant clinical results.In order to explore the clinical effect of Bushen Huoxue Formula in treating unexplained recurrent spontaneous abortion,from the immunological point of view,By observing the expression of IL-1βR and IL-33R in decidual tissues of URSA patients and normal early pregnant women,and by observing the efficacy of 60 patients with unexpained recurrent abortion kideny deficiency and blood stasis syndrome,To investigate the mechanism of Bushen Huoxue formula on IL-1β and IL-33 cytokines,and to provide the theoretical basis for the treatment of unexplained recurrent spontaneous abortion.Method:In this study,subjects were divided into two parts,The first part collects 15 cases of normal early pregnancy were collected decidual tissue as the control group,15 cases of pregnancy failure with unexplained recurrent spontaneous abortion were collected decidual tissue as the study group.The expression of IL-1βR and IL-33RmRNA were detected by RT-PCRmethod in 15 cases of URSA and 15 cases of normal early pregnancy.The second part collected 60 patients(6-8 weeks of pregnancy)with URSA criteria were divided into two groups,30 cases as the study group given Bushen Huoxue formula and Western medicine dydrogesterone treatment,30 cases as the control group were treated with dydrogesterone treatment.Collect their blood 4-5ml when they are in and out of our study.Use ELISA to determine the change of IL-1βand IL-33 level,and to monitor the patients of two groups serum P、E2 and β-HCG levels before and after treatment.Results:1.The expression of IL-1βR in decidua of URSA patients was significantly higher than that of normal group,and there was significant statistical difference(P<0.05);The expression of IL-33R in decidua of URSA patients was significantly lower than that in normal group,and there was significant statistical difference(P<0.05).2.The content of IL-1β in peripheral blood of patients with URSA which as study group and control group was significantly lower than that before treatment,and there was significant statistical difference(P<0.01),The content of IL-33 in peripheral blood of patients with URSA which which as study group and control group was significantly higher than that before treatment,there was significant statistical difference(P<0.01).3.The level of IL-1β in the URSA study group was significantly lower than that in the control group,which after treatment,and there was significant statistical difference(P<0.01);The level of IL-33 in the URSA treatment group was significantly higher than that in the control group,which after treatment,and there was significant statistical difference(P<0.01).4.After treatment for four weeks,The levels of progesterone、estradiol and human Chorionic gonadotropin in peripheral blood of study group and control group were significantly higher than those before treatment,what there was significant statistical difference(P<0.01);The levels of P、E2 andβ-hcg in peripheral blood of study group were higher than control group,there was significant statistical difference(P<0.01).Conclusion:1.Cytokines work by rexeptors,The low expression of IL-1βR and the high expression of IL-33R in decidua tissues of URSA patients,which indicate that IL-1β and IL-33 play differences role in the patients with normal decidual and URSA decidual,which may affect the embryo implantation,development and implantation of embryos,may associated with abortion.2.Bushen Huoxue Formula may improve the clinical efficacy of patients with unexplained recurrent spontaneous abortion by appropriately reducing the expression of IL-1βand increasing the expression of IL-33 in peripheral blood.3.Bushen Huoxue Formula can maintain pregnancy by significantly improve the progesterone,estradiol and specific human chorionic gonadotropin in peripheral blood of URSA patients.
