Objective : Recurrent abortion is a common pathological pregnancy.The incidence rate among couples of childbearing age is 1%-5%,and it is increasing year by year.Among them,the cause of 40% of patients is still unclear,which gives recurrent miscarriage.The prediction,diagnosis,and treatment of the disease have brought difficulties and challenges.It is currently speculated that the reason may be related to the abnormal balance of immune tolerance at the maternal-fetal interface.Therefore,this study used to detect the expression levels of IL-25,IL-17,and IL-33 in the decidua tissue of patients with recurrent miscarriage during the abortion cycle.To compare the expression levels of IL-25,IL-17,and IL-33 in the decidua tissues,and analyze the differences and correlations among early pregnant women,and explore the role of IL-25,IL-17,and IL-33 in the decidua tissues in recurrent miscarriage.The role and related effects of the patient’s maternal-fetal interface immune tolerance balance regulation,and thus lay a certain theoretical foundation for the study of the mechanism of cytokines regulating the maternal-fetal immune tolerance balance in recurrent abortion.Method:Strictly follow the inclusion and exclusion criteria to randomly select a case group of women who underwent uterine evacuation after recurrent spontaneous abortion in the Department of Gynecology of Sichuan Provincial People’s Hospital from December 2019 to December 2020.At the same time,select Normal healthy early-pregnant women whose pregnancy was terminated due to personal factors in the department of gynecology of the Sichuan Provincial People’s Hospital were set as the control group 。 The experiment was approved by the ethics committee of Sichuan Provincial People’s Hospital.All patients adopted the principle of informed consent and signed the informed consent.After the uterine dysfunction,the decidual tissues of the two groups were collected to make tissue homogenates,and the decidual tissues of the patients were detected by ELISA.The levels of IL-25,IL-17 and IL-33 were compared and analyzed.All experimental data were statistically analyzed using SPSS26.0.Result: 1.A total of 84 patients were enrolled in this study,43 of which belonged to the unexplained recurrent miscarriage group(case group),of which 32 were patients with primary recurrent miscarriage and 11 were patients with secondary recurrent miscarriage;41 cases For normal healthy early pregnancy women group(control group);2.There was no significant difference in age,BMI,number of pregnancy,gestational week and APTT between the two groups of patients(p>0.05).The number of deliveries in the case group was significantly lower than that of the control group(p<0.05),and the number of abortions was higher than that of the control group(p <0.05),the difference is statistically significant.3.The levels of IL-25 and IL-33 in the decidua tissue of the case group were lower than those of the control group(p<0.05),while IL-17 was significantly higher than the control group(p<0.05),and the difference was statistically significant.The expression level of IL-25 in the decidua tissue of the case group was not correlated with the number of spontaneous abortions(p>0.05).There was no significant difference in the level of IL-17 in the decidua of patients with 2 spontaneous abortions compared to those with 3 spontaneous abortions(p=0.096),while those with more than 3spontaneous abortions have significantly higher IL-17 levels than those with 2spontaneous abortions,the difference is statistically significant(p=0.012),and those with more than 3 spontaneous abortions are compared with spontaneous abortions The level of IL-17 expression was also significantly increased in 3 cases,the difference was statistically significant(p=0.041);the level of IL-33 in decidua of patients with 3 or more spontaneous abortions was significantly lower than that of 2 spontaneous abortions,The difference was statistically significant(p=0.024,p=0.09),but there was no significant difference in the expression of IL-33 in the decidua of patients with spontaneous abortion 3 times compared with those with more than 3 times(p=0.148).4.There was no significant correlation between IL-25 and IL-17,IL-33,and IL-17 and IL-33 in the decidual tissues of the case group and the control group(p>0.05).Conclusion:1.The expression of IL-25 and IL-33 in the decidua of patients with unexplained recurrent miscarriage was significantly reduced,suggesting that they may be related to the occurrence of unexplained recurrent miscarriage.There is no correlation between IL-25 in decidua tissue and the number of abortions.It indicates that IL-25 is not of high value in predicting the occurrence of spontaneous abortion again.The expression level of IL-33 is different between patients with 2 and 3spontaneous abortions,indicating that IL-33 may have a certain predictive value for the recurrence of spontaneous abortion in unexplained recurrent miscarriage.2.The expression of IL-17 in the decidua of patients with unexplained recurrent miscarriage is abnormally increased,indicating that it may be one of the reasons for the occurrence of URSA,and after more than 2 miscarriages,as the number of miscarriages increases,the expression of IL-17 also Increasingly,this suggests that the expression level of IL-17 in the decidua tissue can be used as a predictor of spontaneous abortion in patients with recurrent abortion.3.There is no significant correlation between IL-25,IL-17 and IL-33 in decidua tissue. |
