Study On The Chemical Constituents And Their Antitumor Metastatic Activities From Rhodiola Wallichiana

Rhodiola wallichiana(HK)S H Fu var Cholaensis(Praeg)S H Fu.grew in the mountain of the height of 3500-5000 meters above sea level,more than 2000 years ago,“Shen Nong Ben Cao Jing","Qian Jin Yi Fang" and "Ben Cao Gang Mu" in the Ming dynasty are recorded.It is distributed in Yunnan,Qinghai and Tibet and other places.Studies have revealed that Rhodiola preparations exhibit adaptogenic effect,including neuroprotective,cardioprotective,anti-fatigue,antidepressive,anxiolytic,nootropic,life-span increasing effects and CNS stimulating activity and so on.Rhodiola contains essential oils,fats,waxes,sterols,glycosides,organic acids(oxalic,citric,malic,gallic,succinic),phenolics including tannins and proteins.By repeated column chromatography,including Silica gel,Toyopearl HW-40,Macroporous resin and preparative HPLC,21 compounds were isolated and purified from the 75%EtOH extract of Rhodiola wallichiana.By analysis of 1D and 2D NMR,UV,IR,as well as MS analysis,the structures of isolated compounds were identified as follows:2-methyl butyric acid-2-O-β-D-glycosidase(1),7-O-syringic-β-D-sedoheptulose(2),trans-rhobupcyanoside A(3),tyrosol(4),syringic acid(5),3-methoxy-trihydroxybenzoic acid(6),scaphopetalone(7),berchemol(8),benzyl alcohol(9),salidroside(10),2,6-dimethoxyacetophenone(11),lotaustralin(12),sec-butyl-O-β-D-glycosidase(13),trihydroxybenzoic acid(14),rhobupcyanoside A(15),glycerol(16),miyaginin(17),chavicol-4-O-β-D-apiofuranosyl-(1→6)-O-β-D-glucopyranoside(18),eugenyol-O-β-D-apiofuranosyl-(1→6)-O-β-D-glucopyranoside(19),methyl gallate(20),gossypetin-7-O-(3"-O-β-D-glucosyl)-α-L-rhamnoside(21).Compound 1-3 are new compounds.Compound 7、8、11、13、16、17、18、19 were isolated from the genus Rhodiola for the first time,compound 12、15、20、21 were isolated from this plant for the first time.The antitumor metastatic activities of these isolated compounds were examined.Compounds 7、10、11、17 showed positive antitumor metastatic activities,and compounds 10 and 17 showed significant antitumor metastatic activities.Many literatures reported chemical structures in the genus of Rhodiola have all kinds of biological activities,but this report first find chemical structures which had anti-tumor metastasis with MD-MBA-231 cell lines.
