Objective:A lot of links were easily led to the loss of effective components of paeoniflorin in Paeoniae Radix Albain in HangZhou,such as the traditional primary processing and processing pieces separately,many intermediate links such as the infiltratin before processing.On the basis of evaluating the suitable harvest period,the preliminary proeessing method,to establish the method and parameter for the origin integration and process in Paeonia Radix Alba by determining the rate of finished products,the contents of water extract,and the quality of pieces with Combined method of determining the content of effective components and Qualitative analysis of HPLC fingerprint.And also the preservation period for fresh was studied by Covering wet sand.Methods:1.We evaluated yield and quality of Paeonia Radix Alba in the different harvest period in terms of fresh weight,dry weight,drying rate,paeoniflorin content,water extract content and so on.2.With the rate of finished products,appearance properties,the content of water extraction and paeoniflorin index,we investigated the quality of Paeonia lactiflora and processed piecesof different primary and integrated processing.3.We built the evaluation method for appraising the quality of Paeonia lactiflora with determination of paeoniflorin content combined with the qualitative analysis of chromatographic fingerprint of HPLC and analyzed the differences between different initial processing and different integration processing technology.4.Through the storage of fresh Paeonia Radix Alba covered with wet sand,we studied the preservation time for it.Results:1.The fresh weight,dry weight,drying rate,the content of peoniflorin and water extracts of Paeonia Radix Alba maintained in a relatively high level from the end of August to early October.Therefore,It is suitable for the harvest period from the end of August to early October.2.In Initial Processing,the way of removing the skin before boiling and steaming was better than removing the skin after boiling and steaming.Among them,the way of removing the skin before boiling was the best and the second is to peel after steaming.Compared with the way of removing the skin before boiling,removing the skin after boiling and steaming would get a higher rate of finished product,but its content of paeoniflorin decreased by 13.20%and 36.40%respectively and its content of water extract decreased by 17.16%and 14.37%respectively.The time of boiling after removing the skin was for 5 minutes appropriately at the 100℃ in the boiling water and the time of steaming after removing the skin was for 10 minutes appropriately.Too long time will reduce the component contents and we pierce the toothpick into the Paeonia Radix Alba as the standard.3.Compared with the methods in traditional processing,the three methods in integrated processing of semi-dry sectioning with baking for 20 minutes at the 100 degrees,semi-dry sectioning with boiling for 5 minutes and semi-dry sectioning with fixing for 6 minutes by microwave after removing the skin were better.And the contents of water extract in these three methods increased by 31.65%,9.85%and 28.94%respectively;the content of paeoniflorin increased by 23.78%,12.36%and 3.50%respectively.Compared with the methods in traditional processing,the two methods in integrated processing of semi-dry sectioning with fixing for 6 minutes by microwave and semi-dry sectioning with steaming for 10 minutes were better.the yield in two methods increased by 11.30%and 8.53%;the content of water extract increased by 38.36%and 9.51%;the content of paeoniflorin increased by 26.20%and 20.01%.4.We established the quality evaluation method with determination of paeoniflorin content combined with the qualitative analysis of chromatographic fingerprint of HPLC.The similarity degre of fingerprints for Paeonia Radix Alba’s different primary processing methods was more than 0.97.In addition to sulfur fumigation,the similarity among the integrated methods were more than 0.98.Compared with the traditional processing,the relative change rate of the ratio of peak area to sample weight increased by 94.7%and 96.1%respectively in the two methods of semi-dry sectioning with baking for 20 minutes at 100℃and semi-dry sectioning with steaming for 10 minutes after removing the skin.And the two processing methods would minimize the loss of effective components in Paeonia Radix Alba.5.The optimum parameter of integrated processing technology with boiling after removing the skin was:took the fresh drug boiled for 5 minutes out of the boiling water,and baked at 60 ℃ to semi-dry(water content 27%-33%),then cut into 3.5-4.0mm Pieces and dried at 60℃.6.The result indicate that preservation time of fresh Paeonia Radix Alba covered with wet sand could last to end of November.And there were only a slight loss in the drying rate and effective components during the storage.Conclusion:1.It is suitable for the harvest period of Paeonia Radix Alba from the end of August to early October;2.Removing the skin before boiling was the best way of initial processing and the second is to peel after steaming.The time of boiling after removing the skin was for 5 minutes appropriately at the 100℃ in the boiling water and the time of steaming after removing the skin was for 10 minutes appropriately.3.the three methods in integrated processing of semi-dry sectioning with baking for 20 minutes at the 100 degrees,semi-dry sectioning with boiling for 10 minutes after removing the skin,and semi-dry sectioning with steaming for 10 minutes were better.The contents of water extract in that increased by 31.65%,9.85%and 9.51%respectively,the content of paeoniflorin increased by 23.78%,12.36%and 20.01%respectively,Compared with the methods in traditional processing.4.The results show that the integrated processing of Paeonia Radix Albain place of production was technically feasible.Not only it can reduce many intermediate links,shorten the processing cycle and save resources,but also help to improve intrinsic quality.
