The Effect Of Warming Kidney And Spleen Recipe On Glomerular Diabetic Nephropathy Key Proteins

Objective:Observe the effect of warming kidney and spleen Recipe on glomerular diabetic nephropathy(DN)key proteins,it provides an experimental basis in clinical application.Methods:The streptozotocin-induced DN 30 rats were randomly divided into five groups,that is model control group,low-dose group(7.5g/kg kidney-warming spleen-fortifying formula),middle-dose group(15g/kg kidney-warming spleen-fortifying formula),high-dose group(30g/kg kidney-warming spleen-fortifying formula)and positive control group(25mg/kg Valsartan),each group has 6 rats,3 rats a cage rearing,each rat was given corresponding drugs once a day,the other 6 rats as normal control group and 3 rats a cage rearing.After 8 weeks treatment,using immunohistochemical and the quantitative fluorescence PCR technology to detection the expression of glomerulus key protein and gene.Result:Compared with the normal control group,the expression of nephrin and podocin which is a protein on sertoli cell of model control group is observably reduce(P<0.01),and the relative transcript level of nephrin and podocin gene is observably promoted(P<0.01).The expression of nephrin and podocin is observably promoted in low-dose group,middle-dose group and high-dose group(P<0.05,P<0.01),the relative transcript level of nephrin and podocin gene is observably reduced(P<0.01).Conclusion:Kidney-warming spleen-fortifying formula can maintain the expression of protein and gene related to sertoli cell approach the level of normal control group.Kidney-warming spleen-fortifying formula may affects the component related to DN rats glomerulus and play a role to protect kidney.
