Objective:To study whether exogenous hydrogen sulfide(H2S)affects burn serum through ATP-sensitive potassium ion channels to intervene in the endoplasmic reticulum stress of epidermal cells.Methods:The neonatal mouse epidermal cells were used as the research object,and they were subcultured to the third generation.The cells in the same culture flask were inoculated into a six-well plate and synchronized for 1 day before grouping.The serum of normal rats and burned rats was prepared by modeling and made into intervention medium.Sodium hydrosulfide(Na HS)and glibenclamide(Gli)were respectively configured into 150μmmol/L and 200μmmol/L solutions for intervention.The experimental groups were:normal group,burn group,burn+Na HS group,burn+Gli group,burn+Gli+Na HS group.Intervene cells according to different intervention conditions.After intervention,q PCR technology was used to detect the expression levels of Caspase-12,CRT,GRP 78,and JNK m RNA in each group of cells;Western Blot technology was used to detect the expression levels of Caspase-12,CRT,GRP 78,and JNK protein in each group of cells.Results:1.The expression results of Caspase-12,CRT,GRP 78 and JNK mRNA in each group were measured under different intervention conditions:(1)Burn group>normal group;(2)Burn group>burn+Na HS group;(3)Burn group+Gli group>burn group;(4)burn+Gli group>burn+Gli+Na HS>burn+Na HS group.2.The expression results of Caspase-12,CRT,GRP 78 and JNK protein in each group were measured under different intervention conditions:(1)Burn group>normal group;(2)Burn group+Gli group>Burn group(3)Burn+Gli group>burn+Gli+Na HS>burn+Na HS group.3.The Caspase-12,CRT and JNK protein expression results of each group were measured under different intervention conditions:burn group>burn+NaHS group.The differences between the above groups were statistically significant,(P<0.05).Conclusion:1.Exogenous hydrogen sulfide can alleviate excessive endoplasmic reticulum stress in epidermal cells interfered by burn serum.2.ATP-sensitive potassium ion channel is the target of epidermal endoplasmic reticulum stress by exogenous hydrogen sulfide protective burn serum intervention.
