Objective: To investigate miR-455 expression variation as well as its impact on GRP78,JNK,GRP78 mRNA,JNK mRNA of burn rat epidermal cell,with intervention of extrinsic hydrogen sulfide.Methods: Rat epidermal cells were subcultured to the third generation and divided into three groups,transfected with miR-455 mimic,miR-455 negative control,miR-455 inhibitor separately.10% serum of burn rat followed by 150μmol/L NaHS was used to intervene experimental groups,After intervention,Western Blot was used to measure GRP78,JNK expression,qPCR was used to measure GRP78 as well as JNK mRNA expression.Results: In miR-455 mimic GRP78,JNK,GRP78 mRNA,JNK mRNA expression was statistically higher(P<0.05),compared with miR-455 negative control.In miR-455 negative control,expression of GRP78,JNK,GRP78 mRNA,JNK mRNA was significantly higher than miR-455 inhibitor(P<0.05).There were statistical differences regarding GRP78,JNK,GRP78 mRNA,JNK mRNA expression between miR-455 mimic and miR-455 inhibitor(P<0.05).Conclusions: With intervention of extrinsic hydrogen sulfide,miR-455 down-regulation could reduce the expression of GRP78,JNK,GRP78 mRNA,JNK mRNA in burn rat epidermal cell,so as to curb ERS.miR-455 exerts protective effect,with hydrogen sulfide intervention,in burn rat epidermal cell.
