Objective:To confirm the expression of miR-409-3p in human keloid tissue and human keloid fibroblasts and its effect on the proliferation,apoptosis,invasion/migration ability and cell cycle of human keloid fibroblasts.Methods:(1)The expression of miR-409-3p in human keloid/normal skin tissue and human keloid/normal skin fibroblasts was detected by qRT-PCR.(2)Cultivating human keloid fibroblasts,divide them into miR-409-3p mimic group and miR-409-3p NC group and transfect with corresponding liposomes.(3)Using EdU to detect cell proliferation ability;Scratch test and Transwell test to detect Cell invasion/migration ability;flow cytometry to detect cell apoptosis and cell cycle.Results:(1)qRT-PCR showed that the expression of miR-409-3p in 8 cases of human keloid tissues was significantly lower than normal human skin tissues(P<0.01)and the expression of miR-409-3p in 5 cases of human keloid fibroblasts was significantly lower than normal human skin fibroblasts(P<0.01).(2)When we overexpressed miR-409-3p in human keloid fibroblasts,the qRT-PCR results showed that compared with miR-409-3p NC group,the expression of miR-409-3p in miR-409-3p mimic group was significantly up-regulated(P<0.001).(3)Compared with miR-409-3p NC group,the cell proliferation(P<0.05)and invasion/migration ability(P<0.001)of miR-409-3p mimic group was significantly decreased and the proportion of apoptosis increased(P<0.001);the proportion of cells in the G1 phase was significantly increased(P<0.01)and the proportion of cells in the S phase was significantly reduced(P<0.001).Conclusion:miR-409-3p is down-regulated in human keloid tissues and human keloid fibroblasts,and up-regulating the expression of miR-409-3p in keloid fibroblasts can significantly inhibit cell proliferation and invasion/migration ability,and can up-regulate the proportion of cells in G1 phase,down-regulate the proportion of cells in S phase and induce apoptosis.It is suggested that miR-409-3p can inhibit the development of keloids by affecting the biological functions of human keloid fibroblasts.
