Objective:Breast cancer has brought great harm to women’s health.It is well known that accurate evaluation to the grade malignancy and metastatic behavior will guide the treatment precisely for an individual patient.In this study,three cancer-testis antigens(CTAs)that melanoma-associate antigen C2(MAGEC2),testes-specific protein 50(TSP50)and Lysine demethylation 5B(KDM5B)were tested in three breast cancer cell lines and cancerous tissues.And the relation of CATs to the clinical characteristics will be analyzed to explore the value for breast cancer molecular typing.The expressive change of TSP50 in breast cancer cells will be detected after applying of genistein(Gen),pyrrolidinedithiocarbamic acid(PDTC)and curcumin(Cur).Methods:The expression of MAGEC2,TSP50 and KDM5B in breast cancer pathological tissues chips and para-carcinoma tissues were detected by method of immunohistochemistry(IHC),and those data were analyzed by SPSS 15.0 software.Three kinds of breast cancer cells that MCF-7,MCF-7/ADR and MDA-MB-231 were cultured on glass coverslips in vitro.The expression of MAGEC2,TSP50 and KDM5B were detected by IHC after the different density of Gen,PDTC and Cur were added to the three kinds of cells for 24 hours.The cells of MCF-7 were disposed by different density of Gen,PDTC and Cur.The cell proliferation inhibition rate(IR)was measured by MTT,and the expressive change of TSP50 in these cells was detected by western blot(WB)assay.Results:The positive expression rate of MAGEC2,TSP50 and KDM5B was 72.22%,64.58%and 62.22%,respectively.MAGEC2,TSP50 and KDM5B were positively related to the tumor size and the TNM staging system.TSP50 and KDM5B were also positively related to N stage.MAGEC2,TSP50 and KDM5B were all expressed on MCF-7、MCF-7/ADR and MDA-MB-231 cell lines.Among of the three kinds of cells,the expressive abundance of TSP50 was the best in MCF-7.The proliferation of MCF-7 and the expression of TSP50 were inhibited by Gen,PDTC and Cur.Conclusions:MAGEC2,TSP50 and KDM5B were positively related to the grade malignancy and metastatic behavior of breast cancer,so it may be used for breast cancer molecular typing.MAGEC2,TSP50 and KDM5B were all expressed in breast cancer.Gen and the inhibitors of NF-κB(PDTC and Cur)could inhibit the proliferation of breast cancer cells by decreasing expression of TSP50.
