Halostachys caspica grows in desert and saline alkali land,which has good ecological value and has certain effects on bacteria-inhibiting.In order to further develop and utilize the application value of Chinese herbal medicine of Halostachys caspica Exploring its therapeutic effects in the clinic.This experiment first isolated and identified S.aureus in milk emulsion of mastitis.The extract of Halostachys caspica and its compound extract were used to test the Bacteriostasis of S.aureus in vitro,To evaluate the irritant and anti-inflammatory effects of Halostachys caspica compound;Through animal infection test,To evaluate the clinical therapeutic effects of Halostachys caspica compound extract on Kunming mice infected with S.aureus.The results provide basic data reference for the development of Halostachys caspica pharmaceutical preparation.1.Seventy-three milk samples of mastitis cows were collected from a large scale dairy farm in Alar City,Xinjiang.All 32 strains of suspected S.aureus were identified by routine bacterial isolation,identification,Gram staining and biochemical tests.16 S r RNA and nuc gene were amplified by PCR.Through sequence comparison analysis,32 strains were identified as S.aureus.Based on 10 virulence factor loci,the virulence genes of 32 strains of S.aureus were detected by PCR.It was found that the types of virulence genes carried by the same strains were different: Among them,The strains carrying TSST gene were the most,accounting for34.3%(11/32),;PVL gene was the least,3.1%(1/32).The isolates carrying different virulence genes were selected to infect mice respectively.The results showed that the more virulence gene types,the stronger the virulence of the isolates.2.Halostachys caspica,dandelion,Pulsatilla and their compound were selected respectively,and the extract was obtained by alcohol extraction,and the concentration of the extract was 1g/ml.According to the different proportion of Halostachys caspica in the compound,the compound was divided into three groups: compound I(30%),compound II(40%)and compound III(50%).In vitro bacteriostatic test was carried out by broth dilution method,and the content in the compound was preliminarily measured,The MIC of compound Ⅰ and compound Ⅲ to S.aureus was 300 mg/m L;The MIC of compound II is 250 mg/m L and the MBC is 250mg/m L.the bacteriostatic effect of compound II is the best.The results of the pharmacodynamic stability test of compound II showed that the antibacterial effect of compound II was not affected by high pressure and the change of p H between(6.0~7.5),and Halostachys caspica compound could be used for clinical treatment after high pressure treatment.3.Kunming mice were divided into 4 groups,including 3 experimental groups was gavaged with 0.4 m L(1g/m L)of Halostachys caspica compound medicine,and the control group was gavaged with 0.4 m L of normal saline.The irritation of yansuimu compound on mouse mucosa was observed by continuous administration.The results showed that there was no difference between the gastrointestinal mucosa of mice in group 1 and group 2and that of mice in the control group,and there was slight congestion on the surface of gastric mucosa in group3.Through the test of xylene induced inflammatory reaction in mice,it was found that the ear swelling rates of Halostachys caspica compound group,Halostachys caspica group,aspirin group and normal saline group were59.22±10.42%,65.45±10.45%,55.71±11.71% and 73.60±10.37% respectively;After the highly pathogenic bacteria isolated in this experiment infected mice,the cure rates of Halostachys caspica compound group and control group were compared.The cure rates of Halostachys caspica compound group and antibiotic group were 80% and 90% respectively,The results showed that Halostachys caspica compound had no irritation to mouse mucous membrane;Halostachys caspica compound also has good anti-inflammatory effect;Halostachys caspica compound also has good therapeutic effect in the treatment of mice infected with S.aureus.The above results showed that the bacteriostatic effects of the compound extract composed of Halostachys caspica,dandelion and Pulsatilla chinensis was better than that of Halostachys caspica and other single herb Chinese medicines.It has no irritation and good anti-inflammatory effect,It has certain effects on Kunming mice infected with S.aureus.The results provide basic reference for the development and utilization of compound preparation of Halostachys caspica.
