| Halostachys caspica is a kind of plant growing in desert and saline-alkali soil,and found that it not only has good nutritional value,but also has a strong antibacterial effect.To further explore it’s application value in medicine and explore it’s clinical medicinal effect,in this study,pathogenic E.coli was identified from lambs with diarrhea in the farm,the bacteriostasis test in vitro was carried out with the single and compositional compound extract of Halostachys Caspica,in vitro bacteriostatic comparison was made between the isolated pathogenic bacteria and the standard strains,the artificial infection cases of clinical pathogenic bacteria were treated clinically,to foundation for the clinical medication of Halostachys Caspica.1.E.coli was isolated and identified from lambs with diarrhea in a large scale sheep farm in southern Xinjiang by PCR and other methods,and the virulence gene and pathogenicity of the identified E.coli were detected.A total of 38 strains of E.coli were isolated and identified from 70 fecal samples of lambs with diarrhea.In the virulence gene detection of 38 isolates of E.coli,it was found that 38 isolates carried different virulence genes: fim C,irp2,hly F,fyu A,with at most four virulence genes and at least one virulence gene simultaneously.The results showed that the strains carrying 4 virulence genes had the strongest pathogenicity,while the strains carrying 1 or 2 virulence genes had the weaker pathogenicity.2.In vitro bacteriostasis experiments were carried out on the crude extracts of Halostachys caspica,Elaeagnus angustifolia,Baitouweng and it’s compound by broth dilution method.The results showed that the antibacterial effect of compound crude extract was better than that of single crude extract medicine.The preliminary results showed that the MIC of compound â… crude extract appeared when the crude concentration was 500 mg/m L,while the minimum MBC was not detected in this experiment and the crude extract of â… had the highest antibacterial potency.The efficacy stability test of compound crude extract â… was carried out.The test results showed that the effect of autoclavic sterilization and the fluctuation of p H(6.0~7.5)had little effect on the antibacterial effect of compound crude extract â… ,which proved that the compound crude extract could be used in clinic after autoclavic sterilization.3.The maximum tolerance test on mice showed that when the crude drug concentration of Halostachys caspica compound â… was 1 g/ m L,the maximum tolerance range was 0.45 m L,but the optimal use range was less than 0.4 m L.It was found that the crude extract of Halostachys caspica compound â… had a certain curative effect in the treatment of animals infected with E.coli.Based on the above experimental results,the main bacterial pathogen of lamb diarrhea in this sampling farm was E.coli.The antimicrobial potency of Halostachys caspica crude extract was not as good as that of the compound extract.Halostachys caspica compound â… of crude extract in animals infected with E.coli has certain curative effect.Therefore,in the development of clinical medicinal use of Halostachys caspica,it can be considered to form a compound for development,so as to improve the clinical efficacy of salinia and better play its medicinal value. |
