Embryo Transfer Technology Research During The Process Of Gene Editing Goat Production

Gene editing is one of the most rapidly developing biotechnology,which is of great significance to accelerate the process of animal breeding.A lot of gene editing animals have been made by using gene editing technology.However,there are still some problems that need to be solved,such as low production efficiency,the key is the connection between gene editing and embryo transfer.In this study,the preparation of gene-edited cashmere goat and Ziwuling black goat was taken as an opportunity to study the matters needing attention in the process of embryo transfer of gene-edited animals.The results showed as follows:1 Superovulation was performed on 82 Shaanbei cashmere goats,all of them were in estrus,the interval of estrus removal was 28.1 ± 8.53 h,and the duration of estrus was31.6 ± 6.45 h.The average number of Corpus luteum was 14.65,and the average number of embryos was 11.72.Among them,the average luteal number,embryo number and embryo recovery rate of the repeated donor goats were lower than those of the first used donor goats.The number of embryos obtained by intubation(P <0.01)and the rate of embryo recovery were significantly better than that by direct method.The effect of the interval time between the last mating and embryo recovery over 20 h was better than that between 15 and 20 h(P <0.05).The number of recovered embryos was higher than that of injected Embryos(P <0.05).2 Superovulated 50 donor Ziwuling black goats,46 were treated effectively,the average number of Luteal was 14.08,the recovery rate of embryo was 79.17%,the average number of Luteal was 10.82.Superovulation method 1 was superior to method 2 in the number of Corpus Luteum,number of recovered embryos and Embryo Recovery Rate(P <0.05).98 Shaanbei White cashmere goats were treated at the same time,and there were 74 effective receptors,including 26 monoluteinized receptors,19 double luteinized receptors,12 triple luteinized receptors and 17 more than four luteinized receptors.The estrus synchronization rate and effective transplantation rate were the highest in the group of Sponge suppository + PMSG 250 unit + domestic FSH 33 unit + PG.There were 74 recipients transferred with an average of 3.56 embryos/goat.After transplantation,15 fetuses were conceived,the transplantation conception rate was 20.27%,the lambing rate was 19,the average lambing rate was 126.67%.The effect of embryo transfer was the best within 16 h from the end of Estrus of donor and recipient to the end of estrus of donor,and the highest in pregnancy rate and lambing rate.3 In this study,51 gene-edited cashmere goats were superovulated,4 were not in Estrus(1 in Group 1,2 in Group 2 and 1 in Group 3),the rest were in estrus normally.Group 3 had the shortest estrus interval(16.44 ± 9.04 H)and the shortest estrus duration(29.33 ± 5.66 h),Group 2 had the largest number of Corpus Luteum(17.18 ± 8.71)and Group 3 obtained the largest number of embryos(8.78).Taken together,it can be concluded that there are many factors affecting the production process of gene-edited goats,which will improve the production efficiency of gene-edited sheep and lay a foundation for the rapid development of animal husbandry in China when the influencing factors were coordinated.
