Paeonia rockii is named because of the spots at the base of the petals.For a long time,P.rockii has been mainly ornamental.In recent years,research has found that the content of unsaturated fatty acids in the seed oil of P.rockii is relatively high,so it is regarded as one.Kind of excellent woody oil crops.However,the oil peony industry is in the early stage of development,and there is a lack of plants with high oil content and stable phenotypes.At the same time,there is also a lack of research on genes related to oil synthesis and seed treatment methods before oil extraction.Therefore,based on the analysis of the phenotypic traits of P.rockii,this study explored whether different traits have an effect on the oil content of P.rockii and whether individual plants with different oil content have differences in gene expression;and explored the changes in the composition and physical and chemical properties of P.rockii seed oil after different treatments,the results show that:1.According to the statistics of phenotypic traits,five individual plants of P.rockii have excellent yield performance,namely ZBMD10,ZBMD12,ZBMD19,ZBMD20,and ZMD22.The yield per plant is greater than 400g/plant;the oil content of the five plants of P.rockii have excellent yield performance,respectively ZBMD5,ZBMD28,ZBMD46,ZBMD48,ZBMD59,and the oil content is greater than 30%.2.According to the comprehensive analysis of phenotypic traits,it was found that the oil content was positively correlated with the outer filament anther ratio,inner filament anther ratio,inner filament length,and outer filament length,but was negatively correlated with the number of fruit angle.3.According to the results of qRT-PCR,it was found that sucrose synthase(SUS),hexokinase(HXK),fructokinase(FPK),fatty acid desaturases 2(FAD2),fatty acid desaturases3(FAD3),acyl-Co A:diacylglycerolacyltransferase(DGAT),phospholipid:diacylgl ycerol acyltransferase(PDAT),oleosins(OLE)genes were expressed at different stages of seed development,and zinc finger protein transcription factors,WRKY transcription factors,b ZIP transcription factors,NFYA transcription factors are also expressed during seed development,and the expression levels of genes and transcription factors in high oil content plants are higher than those in low oil content plants.4.The oil content of P.rockii seeds was the highest(26.32%)after oven drying and shade drying was the lowest(21.22%);there was no significant difference in fatty acid composition,and the content of unsaturated fatty acids was higher.The peony seeds after microwave treatment had the lowest oil content(21.22%),and the content of linolenic acid in seed oil was the highest(424.56mg/g),the lowest(349.63mg/g)after sun drying treatment(P<0.05).5.Different drying methods have an impact on the acid value,peroxide value,saponification value,and iodine value of the peony seed oil.The acid value after shade drying is the highest(6.82mg/g),while the acid value after freeze drying is the lowest(3.1mg/g);the result of peroxide value is shade drying(0.8mmol/kg)>oven drying(0.72mmol/kg)>freeze drying(0.66mmol/kg)>microwave drying(0.65mmol/kg)>sun drying(0.6mmol/kg);the result of saponification value is shade drying(194.72mg/g)>sun drying(183.74mg/g)>oven drying(172.84mg/g)>microwave drying(152.82mg/g)>freezing drying(150.53mg/g);The iodine value of P.rockii seed oil after microwave treatment was higher,165.78g/100 g,and the iodine value of P.rockii seed oil after dry treatment was lower,141.47g/100 g.6.After different drying treatments,there are differences in the total phenol content of peony seed oil.The total phenol content in the peony seed oil after microwave drying is the highest at 22.19mg/100 g,and the total phenol content in the peony seed oil after freeze-drying is lower,17.02mg/100 g.The antioxidant capacity is also different.Comprehensive FRAP,ABTS,DPPH,ORAC antioxidant index analysis shows that peony seed oil has stronger antioxidant capacity after oven drying.
