Chromium(Cr)is a silver-white metallic element with an atomic number of 24.It is widely found in nature.Chromium-containing feed additives are often used in animal husbandry,which can effectively improve the utilization of feed and strengthen the system of livestock and poultry.However,the wrong method will accumulate chromium in the body,posing a serious threat to animals,plants and humans.The liver is one of the main target organs for hexavalent chromium to invade the body.Studies have shown that chromium can cause a series of serious pathological reactions such as oxidative stress and lipid metabolism disorders in liver cells.Nano-selenium is a new elemental form of selenium that has been developed recently.It has the functions of anti-oxidation,immune regulation and anti-aging,and the toxicity of Nano se is lower than that of general inorganic selenium and organic selenium.In terms of acute toxicity,sodium selenite is about 7~22 times of Nano se,and selenium yeast is 4~22 times of Nano se.In order to explore the protective effect of Nano se on hexavalent chromium-induced lipid metabolism damage in LMH cells,the protective function of Nano se was studied starting from the corresponding biochemical indicators and key genes in the regulation of lipid metabolism pathway.The experiment took LMH cells as the object,and treated the cells with potassium dichromate and Nano se alone and together.The test is divided into 4 groups.Control group(C): no addition;Cr(Ⅵ)challenge group(A): potassium dichromate solution 20μmol/L,exposure for 24 hours;Nano se treatment group(T): Add the dose of chromium in the challenge group,and add Nano se 8μg/L after 12 hours;and the Nano se prevention group(P): add the dose of selenium in the treatment group first,and add the dose of chromium in the challenge group after 12 hours,3 repeats in each group.The experiment was performed to observe the morphology of different groups of cells at three time points of 24 h,48h and 72 h after treatment.Use Cell Counting Kit-8 cell counting reagent to detect the proliferation rate,and then measure the biochemical indicators of triglyceride(TG)and low-density lipoprotein sterol(LDL-C),and use RT-q PCR and Western Blot to detect the differential expression of the two genes ACACA and FASN.Explore whether Nano se has a protective effect on chromium-poisoned cells.The results show:(1)Under the influence of Cr(Ⅵ),LMH cell density was significantly reduced,some cell morphology changed,and viability was significantly lower than that of the control group(P < 0.05).The addition of Nano se before and after 12 h of hexavalent chromium treatment of cells can significantly improve the decrease in value-added rate caused by chromium(P<0.05).(2)Chromium causes LMH lipid metabolism damage,which will cause the content of TG and LDL-C to increase.After adding Nano se,the content of both is significantly reduced(P<0.05).(3)After chromium poisoning,The ACACA and FASN in LMH cells are elevated,and Nano se improved the above changes(P<0.05),which confirmed that Nano se can effectively reduce the damage of hexavalent chromium to LMH cells.This experimental results can provide support for the research on the mechanism of chromium-induced animal body damage,and provide new ideas for the development of chromium poisoning protective drugs.
