| Hemerocallis has important edible value,ornamental value and medicinal value.According to statistics,the cultivated area of daylily in the country is about 900,000 mu,and the cultivated area of’Datong huang hua’ in Shanxi province is about 150,000 mu.Based on the genetic map constructed earlier by the research team,this study performed genetic analysis on field traits such as flower diameter,flower bud length,flower bud width,fresh and dry flower bud weight,and calyx height of interspecific and intraspecific hybrid populations.The colchicine and total flavonoids contents in the inter-species hybrid population,as well as the colchicine and vitamin E contents in the intra-species hybrid population were measured and analyzed by QTL.Multi-angle comprehensive analysis and determination of precursor content in the anabolic metabolism process of two kinds of colchicine and colchicine itself were conducted using 32 natural population materials,which laid the foundation for further cloning and fine mapping of important genes in Hemerocallis plants.The main findings are as follows:1.This study established a method for the detection of colchicine anabolic precursors N-deacetylcolchicine and colchicine by high performance liquid chromatography.The method was used to determine 32 hemerocallis materials from natural populations,and the difference among the contents of colchicine synthetic precursors in different flower buds was determined.We measured N-deacetylcolchicine and colchicine at different periods,different processing techniques and different parts of the plant in ’Datong Huanghua’,and finally found that colchicine and N-deacetylcolchicine contents decreased significantly when the bud length was 8 cm,and the contents of N-deacetylcolchicine and colchicine in the leaves were higher than those in the calyx and flower buds.2.Field observations of interspecies hybrid populations were performed during 2018 and 2019.Heritability analysis showed that flower diameter,flower bud length,flower bud width,single fresh bud dry weight,and calyx height in the interspecies hybrid population had heritability of 20.3%-100%.Through the correlation analysis,it is found that the dry weight of a single flower bud has a strong correlation with the dry weight of outside petal and inside petal of a single flower bud.The contents of colchicine in flower buds of parents and F1 population were determined.Based on the field data and QTL mapping for secondary metabolite content,QTL loci related to flower bud length were detected on linkage groups 6,7,and 9 of the interspecific genetic map,respectively.QTL loci associated with flower bud width were detected on linkage groups 1,2,6 and 7.QTL loci associated with fresh weight of a single flower bud were detected on linkage groups 5,7 and 9.QTL loci associated with fresh weight of outside petal of a single flower bud were detected on linked groups 1,5 and 7.QTL loci associated with fresh weight of inside petal of a single flower bud were detected on linkage groups 4,7,and 9.QTL loci associated with the fresh weight of stamens and pistil of a single flower bud were detected on linked groups 4 and 7.QTL loci associated with dry weight of a single flower bud were detected on linkage groups 4,5,6 and 7.QTLs related to the dry weight of the outer petal of bud were detected on linkage groups 3,4,5,6,and 7,QTLs related to the dry weight of the inner petal of bud were detected on linkage groups 5,6,7,and 9,QTLs related to the dry weight of stamen were detected on linkage groups 1,4 and 7.QTLs related to the dry weight of bud and pistil were detected on linkage groups 4 and 7.QTLs related to scape height were detected on linkage groups 1,3,7.QTLs related to the colchicine content of outer petals were detected on linkage group 4 and QTLs were detected on linkage group 7 about the colchicine content of inner petals,the QTL locus related to the content of colchicine in the stamen was detected on linkage group 5.The QTLs associated with the content of pistil colchicine were detected on the linkage group 1,6,7,the QTLs associated with the content of total outer petal flavonoids was detected in 1,5 linkage group,the QTLs associated with the content of total inner petal flavonoids was detected on the linkage group 1,the QTLs associated with the content of total androecium flavonoids was detected on the linkage group 7.The QTLs associated with the content of total pistil flavonoids were detected in the linkage group 2,and the phenotypic explanatory rate of the related traits was 12.8-30.0%.3.The field observations of the in-species hybrid population were conducted during 2018 and 2019.The heritability analysis showed that flower diameter,flower bud length,flower bud width,single fresh bud dry weight,and calyx height in the in-species hybrid population had a heritability of 30%-90%.The highest colchicine content of flower buds in intraspecific hybrid populations was 0.882 μg / g,the lowest was 0.101 μg / g;the highest vitamin E content was 445.05 mg / 100 g,and the lowest was 34.38 mg / 100 g,all of which were super-parental.Based on field data and secondary metabolite content,QTL mapping was carried out.QTL loci related to flower bud length were detected on linkage groups 2,4,7 and 8 in the intraspecific hybrid genetic map.QTL loci associated with flower bud width were detected on linkage group1,2 and 5.QTL loci associated with fresh weight of a single flower bud were detected on linkage group 2,3,7 and 8.QTL loci associated with the dry weight of a single flower bud were detected on the linkage group1,2,3 and 8.QTL loci associated with calyx were detected on the link groups 3 and 10,and QTL loci correlated with the flower bud colchicine content was detected on the link group 1.QTL loci associated with vitamin E content were detected on linkage group 7,and the contribution rate of the related traits was 8.0-21.6%. |
