In this experiment,we studied the suitable plant growth regulator ratio of different cultivars of strawberry,and successfully established the first generation culture of each strawberry cultivar by using the Random District Group experiment to design the variety of the Hongyan,Zhang Ji,Ganlu and Shengdanhong Strawberry as the material.On the basis of the above experiments,the suitable rooting medium for each test variety was screened by using the tissue culture seedling of the female,Zhang Ji and Shengdanhong as the test material.The effects of substrate humidity on the growth of strawberry seedlings were studied by domestication and transplanting of Zhang Ji and mannose-cultured seedlings.At the same time with beauty,Zhang Ji and mannose tissue culture seedlings were used as test materials,and by orthogonal test design,the growth Index and physiological index of strawberry domesticated seedlings with different substrates,different nutrient solution formulas and different trace elements were determined,and then the substrate ratio of the relatively superior cultured seedlings of strawberry test-tube seedlings was screened out.Nutrient solution formula and trace element content combination.The test results are as follows:1.Establishment of vitro culture system of different cultivars of strawberry explants(1)According to the results of swelling number,tufted bud quantity and emergence number of stem tip,the suitable ratio of plant growth regulator with the differentiation of stem tip of Hongyan strawberry is MS+6-BA0.5mg/L+NAA0.2mg/L,and the suitable Zhang Ji of plant growth regulator with the differentiation of stem tip of strawberry is MS+6-BA0.8mg/L+NAA0.2mg/L,suitable for Shengdanhong strawberry,the concentration ratio of plant growth regulator was MS+6-BA0.8mg/L+NAA0.2mg/L,and the concentration ratio of plant growth regulator induced differentiation of sweet strawberry stem tip was MS+6-BA0.8mg/L+NAA0.2mg/L.(2)Carried on the strawberry variety confidante,the rooting test of Zhang Ji and Shengdanhong was used to measure the growth rate,fresh weight,fresh weight,plant height and root length of each variety in rooting culture,and the results showed that the most suitable rooting medium for each variety was 1/2MS+15g sucrose.2.Effect of substrate humidity on transplanting and domestication of different strawberry cultivars in vitroThe growth of the plant was strongest when the moisture content of the substrate was 80%,and the growth of the plants was the most strong when the substrate humidity reached the maximum water holding capacity of the 60%~70%,the plants grew most strong when the moisture in the substrate reached 70% of the maximum water holding capacity of the Zhang Ji.3.Effects of different proportions of matrix,different nutrient solution and trace elements on the transplanting and domestication of different cultivars of strawberry in test-tube plantletsThrough different matrix ratio,a comparative study on the growth,leaf area,plant dry and fresh weight ratio,photosynthetic characteristics,root development index and seedling index of different strawberry seedlings with nutrient solution formula and microelement content was obtained: the most suitable substrate ratio for strawberry cultivation is peat: vermiculite: Perlite =3:1:2,The second is peat: vermiculite: perlite =1:1:1;The nutrient solution formula for strawberry substrate cultivation is: Four water calcium nitrate 236(mg/L),potassium nitrate 353.5(mg/L),two ammonium dihydrogen phosphate(mg/L),magnesium sulfate 123(mg/L),the trace elements of 1/2MS medium had the best effect on strawberry growth.
