Purpose:To explore the effect of Wenjing Decoction on the contents of ASP,Glu,GABA and DA in striatum of TD mice.Material and method:72 ICR mice were randomly divided into 6 groups: normal group,model group,western medicine group,Wenjing Decoction group,split group I(soft liver),split group II(flat liver).In addition to the normal group,the other five groups were injected intraperitoneally with iminodipropionitrile(IDPN)to establish TD model.Indp was injected intraperitoneally at 450 mg /(kg.D)once a day for 10 days.After group intervention,the scores of stereotyped behavior and motor behavior were measured.Enzyme linked immunosorbent assay(ELISA)was used The contents of ASP,Glu,GABA and DA in striatum of mice in each group were measured and the pathological changes of striatum were observed by HE staining.Results:1.Behavioral test: The scores of stereotyped behavior and exercise behavior in the treatment group were significantly lower than those in the model group(P < 0.05);the scores of stereotyped behavior and exercise behavior in the western medicine group and Wenjing Decoction group were not statistically significant(P > 0.05);the scores of stereotyped behavior and exercise behavior in the split group I and split group II were higher than those in the Wenjing Decoction group(P < 0.05).2.Detection of ASP,Glu and GABA in striatum:Compared with the model group,the content of ASP and Glu in striatum of mice in each treatment group decreased significantly(P< 0.05),the content of GABA in striatum of Wenjing Decoction group and split Ⅱ group increased significantly(P < 0.05),the content of GABA in striatum of split Ⅰ group had no statistical significance(P > 0.05);the content of ASP,Glu and GABA in striatum of Western medicine group and Wenjing Decoction group had no statistical significance(P > 0.05),and that of split Ⅰ and split Ⅱ group had no statistical significance(P > 0.05)The content of ASP and Glu in the striatum of group Ⅱ was significantly higher than that of group Ⅱ(P <0.05),and the content of GABA in the body of group Ⅱ was significantly lower than that ofgroup Ⅱ(P < 0.05).Compared with group Ⅰ,the content of ASP and Glu in the striatum of group Ⅱ was significantly lower(P < 0.05).3.DA content in striatum:Compared with the model group,the Da content in the striatum of the other treatment groups was significantly lower(P < 0.05),the Da content in the striatum of the resolution group I was higher than that of the Wenjing Decoction group(P < 0.05),and there was no statistical significance between the western medicine group and the Wenjing Decoction group(P > 0.05)4.He staining pathological changes: The results of Wenjingtang group and Western medicine group were the closest to those of the normal group.Under 400 times of microscope,it was found that the cell morphology was complete,the nucleus was clearly visible,the interstitium was dense and uniform,the neurons were arranged orderly,while the cells in group I and group II were stained deeper,the interstitium was loose and vacuole,and the irregular arrangement of neurons was similar to the results of the model group.Conclusion:1.Wenjing decoction can inhibit the behavioral changes of TD model mice,and can effectively regulate the contents of amino acids(ASP,Glu,GABA)and da.Its effect is similar to that of Western medicine group2.The effect of Wenjing Decoction on reducing the stereotypical behavior and exercise behavior of TD model mice was better than that of group I and group II.3.The effect of Wenjing Decoction on regulating the content of amino acids(ASP,Glu,GABA)was better than that of the two decomposing agents,and the decomposing agent group I had no effect on increasing GABA.4.Wenjing decoction has an advantage in reducing DA content in striatum of TD model mice compared with decomposer group I,while decomposer group II has no effect on reducing DA.
