Objective:To explore the effect of different doses of wenjing decoction on dopamine and D2 receptor sensitivity in striatal rats.Materials and methods:The 72 ICR mice were randomly divided into 6 groups,namely,the normal group,model group,the western medicine group and high dose,medium dose,low dose wenjing decoction group.The mouse model was established for 10 consecutive days with intraPeritoneal injection of 450mg/kgIDPN.Since the building after the completion of the first day,9-11 a.m.daily except normal group and model group to the amount of normal saline,low dose group to wenjing decoction 14 g/(kg·d),the dose group to wenjing decoction 28 g/(kg·d),high dose group to wenjing decoction 56g/(kg·d),the western medicine group to sulfur will benefit hydrochloride 25 mg/((kg·d)for 4 weeks in a row.Time at the end of the fasting,24 h after the treatment,take the brain immediately beheaded,ice bath environment rapid separation of left and right brain,left brain,stripping out the striatum after centrifugation,take that frozen supernatant fluid exist TD-80℃ refrigerator.Set aside.After the right brain is fixed and cleaned,the Paraffin wax block is Preserved at room temperature.ELISA and immunofluorescence were used to detect the experimental indicators,and the results were recorded.ResulTD:Compared with the blank group mice,the behavioral score of the model group was increased(P<0.05).The decrease of behavioral score of western medicine group,high and medium dose group was statistically significant(P<0.05).There was no statistically significant difference between the low-dose group behavioral score and the model group(P>0.05).The behavioral score of western medicine group was not statistically significant compared with the medium dose group(P>0.05).Compared with the western medicine group and the medium dose group,the low dose group behavior score increased(P<0.05).Compared with the high-dose group,the behavioral scores of low-dose group,western medicine group and medium dose group were increased(P<0.05).There was no statistically significant difference between the intracerebral striatum DA in the model group compared with that in the blank group(P>0.05).The content of DA in the brain was lower than that in the model group(P<0.05).The content of DA in the brain of high-dose wenjing tang group was lower than that in the medium dose wenjing soup group(P<0.05).There was no significant difference between the DA content and the model grouP in the low-dose wenjing tang group(P>0.05).In the model group,the fluorescence intensity of DRD2 in the striatum was statistically significant(P<0.05).In the group,high dose of quiet soup striate body DRD2 fluorescence intensity weakened with the model group was statistically significant(P<0.05),high dose grouP of striate body DRD2 fluorescence intensity weakening compared with dose group was statistically significant(P<0.05);There was no significant difference between the DRD2 fluorescence intensity and the model group in the low-dose wenjing tang group(P>0.05).Conclusion:Wenjing decoction can effectively improve the behavioral changes of the mice with the twitching disorder.Wenjing decoction may reduce the dopamine content in the striatum and decrease the high sensitivity of dopamine D2 receptors,thus inhibiting the hyperfunction of neurons,thus achieving the anti-twitching effect. |
