Objective: This topic studies the effects of flavonoids on the bone structure and Wnt3 a and ?-catenin protein expression of rat tibial distraction osteogenesis model,and explores the effective drug concentration of flavonoids on the distraction osteogenesis model in rats.The total flavonoids of Broken Bud provide theoretical basis for promoting bone formation and repair in distraction osteogenesis,accelerating osteogenesis rate,and improving osteogenesis quality.Methods:The left tibia of 60 male SD rats was osteotomized and shortened by 4mm,and fixed with a ring external fixator.After the model was established successfully,it was stratified according to body weight and applied random number table method after 1 week of adaptive feeding.Grouped into high-dose group,middle-dose group,low-dose group and model group,15 rats in each group were given flavonoids of Gusuibu for experimental intervention by intragastric administration.After the model began to intervene throughout the process.All the experimental animals were taken for 30 days after continuous administration.The samples were taken for gross observation and X-ray observation to observe the healing of the osteotomy site and Western Blot detection of Wnt3 a and ?-cantenin protein expression.Results:1.All rats can tolerate the operation and stretching process well,and can move normally without death;60 male SD rats are stable and effective after external fixation;2.X-ray examination:high-dose,middle-dose group and low-dose group have high bone density,the density is close to the normal bone density,and the boundary of the stretch gap is blurred.In the model group,the callus was incompletely filled and there was a low density shadow in the interstitial area.3.Wnt3 a and ?-cantenin protein expression:Wnt3a and ?-canteninprotein expression levels were higher in the high-dose group,middle-dose group,and low-dose group than the model group(P<0.05);Wnt3a and ?-cantenin protein expression in the middle-dose group The level was higher than the low-dose group(P<0.05);the high-dose group Wnt3 a and ?-cantenin protein expression level was higher than the low-dose group(P<0.05);the medium-dose group and the high-dose group Wnt3 a,?-cantenin protein expression level Significance(P>0.05);Conclusion:Total flavonoids of Bupleurum can increase the expression of Wnt3 a and ?-cantenin protein in rat tibial distraction osteogenesis model,and promote the osteogenesis and calcification of rat distraction osteogenesis model.
