Objective:To investigate a novel restoration method of osteogenesis for alveolar defect and other osseous defect through periosteal distraction in a rabbit model. In this study, we were to applicate Ad-hBMP-2 topically on the basis of the established periosteal distraction animal model in rabbits and to detect the action in bone regeneration of periosteal distraction, in order that we might find a new way to improve bone regeneration of periosteal distraction. Methods:1. A self-made periosteal distraction device was rigidly fixed to the lateral surface of the mandible in 16 mature male New Zealand white rabbits. The periosteum were fixed in situ for 7 days after operation. Periosteum was distracted by 2mm from the surface of the mandible at 8th day postoperatively. During the next 20 days periosteum was further distracted by 1mm every 4 days. Finally, the periosteum was distarted 7mm from the surface of the mandible. The unoperated, contralateral side of the mandible served as the control. Four rabbits were killed randomly at postoperative days 28, 35, 42, and 56. The specimens were studied by X-ray, histomorphology and histomorphometrical analysis.2. Thirty-two mature male New Zealand white rabbits were randomized into two groups of 16 rabbits each.
