The Effect Of Qufeng Huoxue Pills On The Expression Of Notch1, Notch3 And Jagged-1 In The Retina Of EAU Rats

Objective:By establishing IRBP to induce the experimental autoimmune uveitis(EAU)rats model,observe the effect of Qufeng Huoxue Pill on the expression of Notchl,Notch3 and Jagged-1 on the EAU of retina on rats,explore the therapeutic effect and mechanism of Qufeng Huoxue Pill on EAUMethods:48 SPF pure male Lewis rats(96 eyes)were divided into a blank group and a model according to random digital table method.The model was established under the IRBP induction to establish an EAU rat model.After successful modeling,they were randomly selected as model control group,Qufeng Huoxue Pill low-power group and Qufeng Huoxue Pill high-power group according to random digital table method.After immunization,the ocular inflammatory reaction was observed under slit lamp microscope every day,and the inflammation score was evaluated according to the Caspi scoring standard.On the 7th day after the establishment of the model,3 EAU rats and 1 blank group of rats were randomly selected and killed,and then their eyeballs were taken to make pathological sections for histopathological examination.After immunization,the Qufeng Huoxue Pills low-power group and the Qufeng Huoxue Pills high-power group were given intragastric administration of Qufeng Huoxue Pills 4g/kg/d and 8g/kg/d for 14 days,respectively,and the model group and the blank group were given intragastric administration of distilled water.After 14 days of oral administration,the relative expression of Notch1 mRNA,Notch3 mRNA and Jagged-1 mRNA in retinal tissue was detected by qPCR method,and the relative expression of Notch1,Notch3 and Jagged-1 protein in retinal tissue was detected by Western Blot method.Results:1.The EAU rat model was successfully established.2.The results of real-time fluorescence quantitative PCR for Notchl,Notch3,and Jagged-1 showed that the mRNA expression of Notchl and Notch3 in the model group was higher than that in the blank group and the intervention group of Qufeng Huoxue Pills(P<0.01),and the expression of Jagged-1 mRNA in the model group was lower than that in the blank group.In the intervention group of Qufeng Huoxue Pills(P<0.01);the expression of Notch1,Notch3,and Jagged-1 mRNA in the low power group and the high power group of Qufeng Huoxue Pill were not significantly different(P>0.05);The protein expression of Notchl and Notch3 was higher in the blank group and the Qufenghuoxue pill intervention group(P<0.01),and the Jagged-1 protein expression in the model group was higher than the blank group than the Qufenghuoxue pill intervention group(P<0.01).Noch1,Notch3,Jagged-1 protein expression was not significantly different between the pill low power group and the high power group(P>0.05).Conclusion:1.By actively immunizing Lewis rats with IRBP antigen-complete Freund’s adjuvant,an EAU rat model can be successfully established.2.Qufeng Huoxue Pills may affect autoimmune uveitis by regulating Notch signaling pathway.
