The Study Of Shoutaiwan On Gene Expression Of AQP6 And AQP12 In Decidua Tissue Of RSA Rats

Objective: RT-PCR was used to detect the expression of aqp6 and aqp12 in rats with recurrent abortion.The effect of shoufetal Pill on aqp6 and aqp12 was clarified.The mechanism of shoufetal Pill on recurrent abortion was discussed from the perspective of kidney water and liquid transport.Methods: To establish a rat model of kidney deficiency and luteal inhibition of recurrent abortion,90 SPF grade SD rats were selected after quarantine for one week.Among them: female rats: 60,male rats: 30,according to the random classification table,60 female rats and 30 male rats were mated in a cage according to the ratio of 2:1 male to female.In the morning of the next day,if the vagina plug of the female rat is off and a large number of sperm are seen under the microscope,it is regarded as the 1 th day of pregnancy.Then 60 pregnant females were divided into 6 groups,10 in each group,and each group was labeled.The modeling time was 10 days in total,and the drugs were given at 9:00 and 16:00 respectively;according to the body weight of female rats,the concentration of the preparation solution was as follows: from the first day to the ninth day of pregnancy: in the morning,the drugs in each group were: group A was given high-dose shoufetal pills(2.0g / kg),group B was given medium dose shoufetal pills(1.0g / kg),group C was given low-dose shoufetal pills(0.5g / kg),group D was given Diqu Progesterone tablet(3.02 mg / kg)e and f groups were given2 ml of normal saline each;in the afternoon,a,B,C,D and E5 groups were given 450 mg / kg of hydroxyurea,and in the afternoon,F group was given2 ml of normal saline each;thus,the weight and diet of female rats were measured on the 1st,4th and 9th day of pregnancy;at 9:00 am on the 10 th day of pregnancy,except F group(normal control group),the other groups were given mifepristone Strychnine tablets(4mg / kg).All pregnant rats were anesthetized with 10% chloral hydrate solution on the 11 th day of pregnancy and then dissected.The Decidua Tissue of uterus was scraped and stored in a refrigerator at-80 ℃ for RT-PCR detection.Results: Shoutaiwan can improve the hair,mental state,activity,diet,weight and other general conditions of rats with recurrent abortion,as well as the condition of blood stasis in uterus and the change of slub like.RT-PCR showed that Shoutaiwan could up regulate AQP6,but had no significant effect on AQP12.Conclusion:Shoutaiwan may up regulate AQP6,a water channel protein,to affect the balance of water metabolism at the maternal fetal interface,so as to intervene the treatment of recurrent abortion,while Shoutai Pill has no significant effect on the regulation of AQP12.
