ObjectiveTo study Shoutaiwan on recurrent abortion model rat uterine decidua tissues of various water channel protein expression and the effect of preliminary screening of water channel proteins associated with recurrent abortion type,its possible mechanism of fetus,expounds life of tires pill Ann functions and the material basis of "kidney advocate water" prevention and treatment of recurrent abortion for traditional Chinese medicine to provide new research ideas.MethodsAdaptability to feed healthy SPF SD rats after one week,female and male mice according to the proportion of 2:1 and cage for the night,and in the morning watch vaginal suppository falls off and vaginal smears under a microscope to see a lot of sperm to determine 0days for pregnancy,in order of pregnancy were randomly divided into life of fetal pill for high,medium and low dose group,flexor progesterone group,model group and normal pregnancy group.Except normal pregnancy group and other groups in pregnancy pregnant rats of day 1 to day 9,every afternoon to hydroxyurea piece of aqueous lavage(450 mg/Kg),in pregnancy day 10 morning with mifepristone tablets lavage solution(4.0 mg/Kg).In addition,in the day 1 to day9 of the pregnancy,the pregnant rats in the high,medium and low dose group were given a daily dose of traditional Chinese medicine(2.0g/Kg,1.0g/Kg,0.5g/Kg)respectively.The pregnant rats in the diprogesterone group were given diprogesterone aqueous solution(3.02mg/Kg)every morning.The model group and the normal pregnant group were given an equal amount of 0.9% sodium chloride injection everymorning.Both groups of pregnant rats from the last 24 hours after dosing broken neck death,aseptic conditions anatomy of pregnant rats take its decidual tissue of the uterus,protein imprinting method is used to inspect each pregnant rat uterine decidua tissue water channel protein expression.Results1.Fetal development in each group.Model group of pregnant rat uterus is black,dark red in colour appearance of the uterus changes like bamboo samples,and embryonic form size in homogeneity,anatomy of the uterus can be found with black blood stasis points,significant reduction of embryo implantation or are already machine embryonic organization.Normal pregnancy pregnant rat uterus appears reddish,found no change bamboo samples,embryonic form relatively uniform size,cut open the womb found no obvious blood stasis points,significant reduction of embryo implantation points or are already machine embryonic organization.Life of fetal pill for high,medium and low dose group,flexor progesterone pregnant rat uterus appears pink or dark red,uterine appearance did not change obviously bamboo samples,size of embryonic form a uniform or non-uniform,dissect uterus uterus not seen blood stasis points,or I can be found that blood stasis points,not seen significant reduction of embryonic tissue of embryonic implantation point or machine.2.Comparison of abortion rates among different groups of pregnant rats.Compared with the pregnant rats in normal pregnancy group,the abortion rate of pregnant rats in the model group,the progesterone group,and the high-medium and low-dose group was significantly increased,and the difference was statistically significant(P<0.05).After drug intervention,compared with model group,the life of fetal pill in high,medium and low dose group and flexor progesterone group of pregnant rats abortion rate significantly decreased,the difference was statistically significant(P<0.05),the life of tire bolus dose group,the life of fetal pill in high dose group and to bend progesterone pregnant rats abortion rate than there were no statistically significant difference(P>0.05).3.Comparison of the expression of water channel proteins in the deciduated membranes of pregnant rats in each group.Compared with the pregnant rats in normal pregnancy group,the AQP0 expression was highly expressed in the model group,and the difference was statistically significant(P < 0.05).After the drug intervention,the expression level of AQP0 in the decidua group was lower than that in the model group,and the difference was statistically significant(P<0.05).Compared with the pregnant rats in normal pregnancy group,the low expression of AQP1、AQP2、AQP3、AQP4、AQP5、AQP6、AQP8、AQP9、AQP10and AQP11 in the model group was statistically significant(P<0.05).After drug intervention,compared with model group,the life of fetal pill in high,medium and low dose group and flexor progesterone group can make pregnant mice decidual tissue AQP1、AQP2、AQP3、AQP4、AQP5、AQP6、AQP8、AQP9、AQP10、and AQP11 expression levels,the difference was statistically significant(P<0.05).Compared with the pregnant rats in normal pregnancy group,AQP7 showed low expression in the decidual tissue of the model group,and the difference was not statistically significant(P>0.05).Compared with the pregnant rats in normal pregnancy group,AQP12 was highly expressed in the decidual tissue of the model group,and the difference was statistically significant(P<0.01).After the drug intervention,compared with the model group,the high,medium,low dose group and the diprogesterone group failed to reduce the expression of AQP12 expression in the decidual tissue of pregnant rats.ConclusionThis study shows that life of fetal pill can effectively reduce the recurrent abortion of pregnant rats embryo absorption rate,promote embryonic development,can cut recurrent miscarriage AQP0 expression in rat uterine decidua tissue,at the same time raise AQP1,AQP2,AQP3,AQP4,AQP5,AQP6,AQP8,AQP9,AQP10,AQP11 protein expression,AQP instructions may be micro material basis of "kidney advocate water" life of fetal pill may be by adjusting the water channel protein expression and play a role of fetus.
