| Objective: By studying the effect of Wandai Decoction on pelvic inflammatory disease,measuring the concentration changes of tumor necrosis factor-α(TNF-α)and transforming growth factor-β1(TGF-β1)to explore the immunological mechanism of Wandai Tang in treating pelvic inflammatory disease;The effect "gold index" changes in the organization’s pharmacology verify the strength of its pharmacological effect.Methods:In this experiment,SD female rats were used as test subjects,46 female rats were randomly selected according to the random number table method,and 10 blank groups were randomly selected,and the sequelae model of pelvic inflammatory disease was replicated by "mechanical injury plus mixed bacteria infection modeling method" After successful modeling,they are randomly divided into model group,Wandaitang group,and antibiotic group by random number method;using HE staining method,Observe the morphological changes of the uterus under light microscope;immunohistochemical method was used to detect the changes in the concentration of tumor necrosis factor-α(TNF-α)and transforming growth factor-β1(TGF-β1).Results:Compared with the normal group and the model group,P<0.05,the difference was statistically significant,indicating that the rat model of pelvic inflammatory disease sequelae was successful.Compared with the normal group,the antibiotic group,P>0.05,the difference was not statistically significant,it could not explain that the concentration of TGF-β1 and TNF-α in the serum of the antibiotic group rats was different from the normal group;the Wandaitang group was compared with the normal group,P>0.05,the difference was not statistically significant,and it could not explain that the concentration of TGF-β1 and TNF-α in the serum of rats in Wandaitang group was different from that in normal group.Comparing the Wandaitang group with the antibiotic group,P> 0.05,the difference was not statistically significant,indicating that the effects of the two drugs on serum TGF-β1 and TNF-α concentrations in rats with sequelae of pelvic inflammatory disease may be equivalent.Conclusion:1.The SPID rat model can be successfully prepared by mechanical injury plus mixed bacterial infection method,and its pathogenic mechanism is similar to that of the human body,but this method requires a higher level of operation by the experimenter,and the mortality rate of rats compared with other modeling methods may be higher.The serum concentrations of TGF-β1 and TNF-α in the model group and Wandaitang group were significantly different.2.The model group’s uterine histopathology often shows a lot of degeneration and necrosis of epithelial cells,gland loss and a large number of inflammatory cell infiltration,uterine smooth muscle fiber hyperplasia and other manifestations.Although antibiotics can improve the inflammatory symptoms in rats,its The repairing effect of pathological changes in rat uterine tissue is not as good as Wandai Decoction.3.Wandai Tang may play a therapeutic role by inhibiting the expression of TGF-β1 and TNF-α protein in rats and reducing the body’s inflammatory response.4.Wandai Decoction and antibiotics may be equivalent in the clinical treatment of SPID.Although they can inhibit the expression of TGF-β1 and TNF-α protein,from the perspective of histopathological changes in rats,their repairing effect on pelvic environment Better than antibiotics.In recent years,the abuse of antibiotics has caused some pathogenic bacteria to become resistant to drugs.However,they cannot achieve better therapeutic purposes.Although they can improve some clinical symptoms,they are prone to relapse.Repeated application of antibacterial drugs can also easily damage the spleen and stomach,causing the body’s immunity to decline.At present,the scope of use of antibiotics is shrinking,and the use of Chinese medicine Wandai Tang can provide more choices for clinical medicine. |
