Rhizoma cyperi(Xiangfu)is the dry rhizome of the Cyperaceae sedge(Cyperus rotundus Linn.sp),its nature is flat,spicy,slightly bitter,sweet,into the liver,three coke meridians.It has the functions of regulating qi and relieving depression,regulating menstruation and relieving pain.It is widely used for liver qi stagnation,chest,flank,abdominal pain,indigestion,irregular menstruation,menstrual dysmenorrhea,cold and abdominal pain,breast pain and many other diseases.The Cyperaceae mainly contains quinones,flavonoids,triterpenes and sterols,volatile oils,sugars,alkaloids,phenols and other chemical components.Modern pharmacological studies have shown that Xiangfu has many biological activities such as anti-dysmenorrhea,anti-mutation,anti-oxidation and blood fat reduction,anti-thrombosis,anti-bacterial,anti-inflammatory and sedative.In recent years,the volatile oil of Xiangfu has received wide attention because of its good physiological activity.At present,the yield of Xiangfu volatile oil is not satisfied and the composition of related products in the market is quite different.Based on this,this article uses Xiangfu as a raw material medicine,optimized the extraction process of volatile oil from Xiangfu,analyzed the main components of volatile oil from Xiangfu and studied their in vitro activities.In addition,since the volatile oil is a kind of oral poorly soluble oil,the traditional oral administration method has low bioavailability in the water-soluble environment in the body,and may cause gastrointestinal irritation,resulting in a series of adverse reactions.Therefore,this topic through the self-emulsifying drug release system,prepared the fragrant volatile oil into a self-emulsifying preparation,in order to improve its bioavailability and reduce adverse reactions.The research results are as follows:(1)The volatile oil of Rhizoma cyperi is extracted by steam distillation.Through the response surface optimization software,the effects of the extraction time,the ratio of material to liquid and the particle size of the drug on the oil yield were analyzed and compared.The results showed that the extraction time had the greatest influence on the oil yield of the medicinal materials,the granularity of the medicinal material has a decisive influence on the difficulty of cleaning the instrument after the medicinal material is extracted.Through optimization,when the optimal process is the extraction time is 5.78 h,the liquid-to-liquid ratio is 4.07:1,and the particle size of the medicinal material is 0.51 mm,the highest oil yield is predicted。Taking into account the actual operation,the extraction time was set to 5.8 h,the liquid-to-material ratio was set to 4:1,and the medicinal material granularity was controlled at 0.5 mm.Under this condition,the actual measured oil extraction rate can reach 0.78%,which is close to the predicted value.(2)The GC-MS combined technology was used to compare the components of the volatile oil obtained in the previous period with the volatile oil of supercritical extraction.The test results were compared with the standard spectrum.The volatile oil extracted from water identified 37 active ingredients,the main components of which were aristolochone(17.08%)and a-Gurjunene(14.54%);supercritical extraction volatile oil identified 35 active ingredients,the main components of which are Isolongifolene(14.18%)and methyl Methyl ricinoleate(13.43%).a-Cyperone and a-Gurjunene are both contained in two oils,but their content is different:the volatile oil extracted by water is 7.10%and 14.54%,respectively;the supercritical extraction volatile oil is 0.65%and 6.14%,respectively.(3)The in vitro antibacterial activity of volatile oil from Xiangfu was investigated by filter paper diffusion method.The results showed that the volatile oil of Xiangfu showed good inhibition to Staphylococcus aureus and Pseudomonas aeruginosa.The DPPH free radical scavenging method,ABTS free radical scavenging method and FARP method were used to investigate the in vitro antioxidant activity of the volatile oil of Xiangfu.The results showed that it had certain scavenging ability to DPPH free radicals and ABTS free radicals,and showed good Restore ability on Fe3+.(4)Using self-emulsifying drug delivery systems(SEDDS)as carriers,Through solubility experiment,orthogonal optimization and ternary phase diagram drawing method,the in vitro emulsification effect,emulsion equilibrium time,emulsion particle size and distribution are used as evaluation indexes,the best prescription and ratio for screening are fragrant volatile oil:Tween 85:absolute ethanol 25:34:41.The formula is matched to perform a series of stability experiments such as stored inhigh temperature,low temperature,strong light,alternating hot and cold,normal temperature conditions for a long while.By observing the particle size,distribution,determination of zeta potential,self-emulsification speed and other indicators,it is found The fragrant volatile oil SEDDS is very stable.Therefore,the research results can provide a scientific basis for the clinical rational application of volatile oil. |
