Antioxidant Effect Of Traditional Chinese Medicine External Application Ointment On Nrf2 In AGA Rats

Purpose:The acute gouty arthritis model mice were established by intra-articular injection of MUS solution.The effects of external application of traditional Chinese medicine on the antioxidant activity of Keap1-Nrf2-ARE signaling pathway were investigated.The swelling degree of ankle joint in AGA model mice was observed.The activities of antioxidant enzymes(SOD,CAT,GSH-Px)in serum and the expression of Nrf2 in synovium were also observed.Material and method:1.Forty SPF SD rats were randomly divided into five groups: normal group(A),model group(B),traditional Chinese medicine external application ointment group(C),colchicine group(D),and rotaline ointment group(E),with 8 rats in each group.The experiment was carried out 7 days after adaptive feeding in the laboratory.The group of external application of traditional Chinese medicine and the group of Futalin ointment were given external application of ointment twice a day.The group of colchicine was given intragastric administration of colchicine,0.3 mg/kg/d.The normal group and the model group were given intragastric administration of normal saline for 5 days.On the third day,except for the normal group,the right ankle joint of the model rats was made by intra-articular injection of MUS solution,and the normal group was made by intra-articular injection of saline.2.The rats were killed by anesthesia one hour after the last gastric perfusion.The activities of SOD,CAT and GSH-Px in serum of model rats were determined by colorimetry,and the average optical density of Nrf2 expression in synovial tissue of ankle joint of rats was determined by immunohistochemistry.Results:1.Compared with the blank group,there were significant statistical differences(P <0.01)in each group 2 hours,6 hours,20 hours and 48 hours after modeling.Compared with the model group,there were statistical differences in colchicine group and traditional Chinese medicine external application ointment group(P < 0.05),butthere was no statistical difference in Futalin ointment group(P > 0.05).2.Compared with the blank group,SOD activity in serum of rats in the other groups had significant difference(P < 0.01).Compared with the model group,SOD activity in the three treatment groups had significant difference(P < 0.01).There was no significant difference among the three treatment groups(P > 0.05).The activities of CAT in serum of rats in the other groups were significantly different from those in the blank group(P < 0.01).Compared with the model group,the activities of CAT in the three treatment groups were significantly different(P < 0.01).Compared with the raditional Chinese medicine external application ointment group,the activities of CAT in serum of rats was no significantly different(P > 0.05).There was significant difference between the colchicine group and the Futalin ointment group(P <0.05).The activities of GSH-Px in serum of rats in the other groups were significantly different from those in the blank group(P < 0.01).Compared with the model group,the activities of GSH-Px in the three treatment groups were significantly different(P< 0.01).Compared with the raditional Chinese medicine external application ointment group,the activities of GSH-Px in serum of rats was no significantly different(P >0.05).There was significant difference between the colchicine group and the Futalin ointment group(P < 0.05).3.Compared with the blank group,the average optical density of Nrf2 positive staining area in the right ankle synovial tissue of rats in model group,traditional Chinese medicine external application ointment group,colchicine group and Futalin ointment group increased significantly(P < 0.01);compared with model group,the average optical density of Nrf2 positive staining area in the right ankle synovial tissue of rats in traditional Chinese medicine external application ointment group,colchicine group and Futalin group increased significantly(P < 0.05).The average optical density of Nrf2 positive staining area in synovial tissue of right ankle joint of rats in ointment group increased significantly(P < 0.05),but there was no significant difference among the three treatment groups(P > 0.05).Conclusion:1.The rat model of acute gouty arthritis was induced by sodium urate crystallization,and the external application of Chinese herbal medicine ointment could significantly reduce the swelling of ankle joint in gouty arthritis rats.2.Traditional Chinese medicine external application ointment can improve the activity of antioxidant enzymes SOD,CAT,GSH-Px in rats with acute gouty arthritis,play an antioxidant role,prevent and treat the tissue and cell damage in rats with acute gouty arthritis.3.External application of traditional Chinese medicine ointment can increase the expression level of Nrf2 in rats with acute gouty arthritis,promote the activation of Nrf2-ARE pathway and increase the production of antioxidant enzymes,so as to achieve the antioxidant effect in rats with acute gouty arthritis.
