| ObjectiveObserve the effect of the cluster needling of scalp acupuncture on neurological function score and expression of protein Cdc42 and GFAP in different time points in rats with acute focal cerebral ischemia.Explore the possible mechanism of scalp acupuncture treatment for ischemic stroke.Methods54 healthy clean-grade male SD rats were randomly divided into three groups,named the false operation group(A,n=18),model group(B,n=18),and the cluster needling of scalp acupuncture group(C,n=18).Every group was divided into three groups(6 rats in each group)according to time points of 24h,7d,14d.The local cerebral ischemia rats model in Group B and C were prepared by modified suture-occluded method,while Group A was sham model.Rats in Group C were treated by the cluster needling of scalp acupuncture,by contrast,Group A and B were intervened without acupuncture treatment.The rats in each group were evaluated by Zea-Longa’s scoring system after being operated and being killed.Detect and identify the expression of protein Cdc42 and GFAP in the brain tissues of MCAO rats in each time point by LFQ proteomics methods.Observe the effect of the cluster needling of scalp acupuncture on the expression of protein Cdc42 and GFAP,and explore the possible mechanism of scalp acupuncture treatment for ischemic stroke.Results1.Neurological scoreIntra-group comparison:The neurological score in group C and B 4h after the operation compared with that in group A was statistically significant different(p<0.05).There was no significant difference between group C and group B(p>0.05).The neurological score in group C and Bcompared with that in group A in all time pionts was no significant difference(p<0.05).In the point of 24h,there was no significant difference between group C and group B(p>0.05).However,in the point of 7d and 14d,the neurological score in group C was lower than that in group B,there is a statistically significantdifference(p<0.05).Inter-group comparison:In the group B,the neurological score at the time point of 14d was lower than that at the 24h and 7d,the three groups didn’t show any statistically difference(p>0.05).In group C,the neurological score at the time point of 14d was lower than that at the time point of 24h2also there was a statistically significant difference(p<0.05).We can found that there was no statistically difference(p>0.05)of theneurological score between the time point of 14d and the group of 7d.There was no statistically difference(p>0.05)of theneurological score between the time point of 7d and 24h.2.Expression of protein Cdc42 and GFAPExpression of protein Cdc42In the 24hpoint,the expression of Cdc42protein in model group was lower than that of false operation group(p<0.05).The expression of Cdc42protein was up-regulated significantly compared with the model group(p<0.05).There was no difference that expression of Cdc42protein in all groupsin 7d and 14d points.Expression of proteinGFAPIn the 24hpoint,the expression of GFAPprotein in model group was lower than that of false operation group(p<0.05).The expression of GFAPprotein was up-regulated significantly compared with the model group(p<0.05).In 7d and 14d point,the expression of GFAPprotein in model group was higher than that of false operation group(p<0.05).The expression of GFAP in acupuncture group was significantly higher than that in model group(p<0.05).Conclusion1.The method of cluster needling of scalp acupuncture can obviously reduce the score of nerve function in MCAO rats,improve the motor dysfunction of rats,and promote the recovery of neural function in rats with cerebral ischemia.2.The expression of Cdc42 protein was significantly decreased after acute cerebral ischemia,and increased after the interviention of cluster needling of scalp acupuncture.The cluster needling of scalp acupuncture can significantly up-regulate the expression of Cdc42 protein in ischemic brain tissue.It may regulate the division and migration of nerve cells,promote the axon repair and regeneration of injured neurons,alleviate the nerve damage.3.The expression of GFAP protein was significantly decreased after acute cerebral ischemia,and increased after the interviention of cluster needling of scalp acupuncture.The cluster needling of scalp acupuncture can significantly up-regulate the expression of GFAP protein in ischemic brain tissue,improve the activity of astrocytes and enhance the tolerance of neurons to play a neuroprotective role after cerebral ischemia. |
