| Background:In recent years,the incidence of hyperuricemia(HUA)has been increasing year by year.The high age is middle-aged and elderly men and postmenopausal women.However,in recent years,there has been a trend of rejuvenation,which has brought heavy burdens to society and families.The treatment of HUA has become a research focus and hot spot in the field of medicine,but neither traditional Chinese medicine nor Western medicine can be satisfactory.At present,there is still a lot of room for experimental investigation in the treatment of early onset HUAObjective:To investigate the acupuncture effect of the rats in the HUA model through the acupuncture of the rat liver(Liver Shu Point),the original point(Tai Chong)and the kidney(Kidney Shu Point)and the original point(Tai Xi).To observe the serum uric acid(SUA),alkaline phosphatase(ALP),aspartate aminotransferase(AST),carbamide(Urea),and alanine transminase(ALT)in HUA model rats.The effects of the concentration and the changes in the morphology of rat liver cells after acupuncture,and whether there are intervention and special effects of acupuncture points on the HUA model rats,It can provide scientific experimental basis for the acupuncture treatment of HUA in medical clinicMethods:32 healthy male rats were randomly divided into 4 groups.They were blank group,model group,intervention group 1(liver)and intervention group 2(kidney),each group of 8 rats.In the latter three groups of rats,potassium oxazinate 2g/(kg.d)was given as a unit of 10 ml/kg to construct a model of HUA in rats,and acupuncture intervention was performed on each group of acupuncture points;The same volume of saline was given to rats in the blank group once a day.After 10 days,the rat’s posterior orbital vein plexus was taken and the serum uric acid level was measured with a fully automatic biochemical analyzer if it was significantly elevated(P<0.05),can prove the success of the mold;During acupuncture treatment,in order to maintain the stability of the high uric acid index,each group was changed to 1 enema of potassium oxazinate 2g the next day.After acupuncture intervention,each group of rats was used for intraperitoneal inj ection anesthesia with 10%chloraldehyde hydrate(0.3 ml/100g),and then blood was taken from the abdominal aorta(2 to 4 ml)and placed in a centrifugal tube for 1 hour,promoting its solidification.Within 30 minutes,after 4 ℃ 1000r/min centrifugation,take the liquid and put it into a fully automatic biochemical instrument to record the results of each indicator;Then the liver of the rat was stained and treated under an ordinary optical microscope to observe the changes in the internal structure and morphology of the cellsResults:The contents of SUA,ALP,AST,ALT,Urea and the morphological changes of liver cells were detected after the experiment:1.Effect of each group on SUA in rats:Compared with the blank group,the content of SUA(144.67 ± 35.66#)in the model group was significantly increased(P<0.05),with statistical significance;Compared with the model group,the content of SUA(108.02±17.38)in Group 1 and SUA(98.20±8.19)in Group 2 was significantly reduced(P<0.05);2.Effect of each group on ALP in rats:Compared with the blank group,the content of ALP(192.94 ± 38.46#)in the model group was significantly increased and had statistical significance(P<0.05);Compared with the model group,the intervention of 2 groups of ALP(131.87±39.88)decreased(P<0.05);3.Effects of acupuncture on AST,ALT,and Urea in rats:AST,ALT,and Urea were not statistically significant before and after treatment(P>0.05).4.Microscopically observed liver biopsy results in rats(1)In the model group,liver cells in the central vein of the liver lobular were generally shown to have varying degrees of cell edema.The structure of the liver lobular in the group was destroyed.The cells were turbid.Some cells had a small amount of vacuoles in the cytoplasm,and the liver cell cords were not arranged clearly.Radial;(2)The scope of hepatic cell edema in the intervention group 1 was significantly reduced,and the cells were turbid.The structure of the hepatic lobules in the group was relatively incomplete,there were no obvious vacuoles,and the hepatic cell cord arrangement was irregularly radial.The hepatic edema in the 2 groups of interventions was reduced compared to the model group.The structure of the hepatic lobules in the group was relatively completeThere were also a small amount of vacuoles in the cells.The hepatic cells were visible and regularly arrangedResearch conclusion:1.Acupuncture intervention can significantly reduce the content of serum uric acid(SUA),effectively reduce the damage to liver and kidney,successfully intervene in the formation of hyperuricemia(HUA)to a certain extent,and prevent gout.2.Acupuncture intervention can effectively slow down the effect of modelling agent on the morphology of liver cells,slow down the normal erosion process of modelling agent,reduce the degree of damage of liver cells,effectively reduce the damage of liver cells,and interfere with the formation of hyperuricemia(HUA)3.In the therapeutic effect of acupuncture in intervention group 1 and intervention group 2,the effect of acupuncture in intervention group 2 was significantly better than that in intervention group 1 by comparing the content of SUA before and after treatment.To some extent,it was more significant to intervene the formation of hyperuricemia(HUA)and prevent gout.4.Acupuncture in intervention 2 groups had significant effect on reducing the content of ALP in hyperuricemia,but there was no difference in the content of ALP in intervention 1 group,which indicated that ALP was also the result of complex changes in vivo,and the influencing factors of the content change might be not only hyperuricemia. |
