Objective:Performed by blocking cerebral ischemia reperfusion by intravascular embolism(MCAO)model in rats intragastric administration of Huatan Tongluo Decoction on the treatment of rat model,the dynamic changes of inflammatory factors in detection of HMGB1-TLR4-NF-κB signal pathway,brain protective effect of Huatan Tongluo Decoction on MCAO rats.Methods:48 male SPF SD rats were randomly divided into sham operation group(A group),model group(B group),Edaravone Injection group(C group)and Huatan Tongluo Decoction group(D group),to MCAO rat model was made by blocking intravascular embolism,each group were intragastric administration of random sampling 7d,detection and scoring,comparison of rat nerve function defect in brain infarct volume,HMGB1,TLR4,NF-κB expression and inflammatory factor TNF-α、IL-1β、IL-6content.Results:(1)The comparison of the scores of neurological deficit in rats:compared with sham operation group,rats in model group,edaravone group,Huatan Tongluo Decoction group had obvious neurological deficits(P<0.01);compared with the model group,edaravone group and Huatan Tongluo Decoction group after administration of 7d neural function defect score in rats decreased significantly(P<0.01);Compared with the edaravone group,there was no significant difference in the score of nerve function defect in the Huatan Tongluo Decoction group after administration(P>0.05).(2)compare the percentage of the volume of cerebral infarction in rats:compared with sham operation group,model group,edaravone group,Huatan Tongluo Decoction group rat brain infarct volume percentage increased significantly(P<0.01);at 7d after administration,edaravone group and Huatan Tongluo Decoction group rats cerebral infarction volume percentage ratio model group decreased significantly(P<0.01);compared with the edaravone group,there was no significant difference in the percentage of cerebral infarction volume in the rats of the Huatan Tongluo Decoction group(P>0.05).(3)Comparison of expression of HMGB1,TLR4,NF-κB protein in the brain tissue of rats:compared with sham operation group,model group,edaravone group and Huatan Tongluo Decoction group in the rat brain expression of HMGB1,TLR4,NF-κB protein were significantly increased(P<0.01);show that compared with the model group,in the administration after 7d,edaravone group and Huatan Tongluo Decoction group HMGB1,TLR4,NF-κB protein expression were significantly decreased(P<0.01);Compared with the edaravone group,there was no statistical significance in the expression of HMGB1,TLR4 and NF-κB protein in the Huatan Tongluo Decoction group(P>0.05).(4)Rat ipsilateral brain inflammation factorTNF-α、IL-1β.IL-6 content:compared with sham operation group,model group,edaravone group,Huatan Tongluo Decoction group rats with TNF-α、IL-1β、IL-6 were significantly increased in brain tissue(P<0.01);show that compared with the model group edaravone group,and Huatan Tongluo decoction group after administration of 7d,the rat brain inflammation factorTNF-α、IL-1β、IL-6 content decreased significantly(P<0.01);Compared with the edaravone group,there was no significant difference in the contents of inflammatory factors TNF-α、IL-1β、IL-6 in the brain tissue of the Huatan Tongluo Decoction group(P>0.05).Conclusion:The Huatan Tongluo decoction is an anti inflammatory response through the HMGB1-TLR4-NF-κB signaling pathway to protect the brain of MCAO rats. |
