The Effect Of Xiaoshuanyin On TNF-α And IL-10 In Peripheral Blood Of Rabbits With Traumatic Deep Vein Thrombosis

Objective:To investigate the immunological mechanism of Xiaoshuan Decoction on treating deep venous thrombosis by testing the levels of TNF-αand IL-10 at different time after the operation of the model.Methods:50 New Zealand rabbits randomly divide five groups,respectively named the blank group(A group),sham-operated group(B group),saline group(C group)and low molecular heparin sodium group(D group),Xiaoshuan Decoction group(E group).In addition to the blank group and the sham operation group,the rabbit model of deep venous thrombosis was prepared by clamping femoral vein.At the first day after the model was successful,the groups was treated respectively with different medicine.After 1 week of continuous treatment,the blood of abdominal aorta was taken after 4h of the first,third and 7d after surgery,and the levels of TNF-αand IL-10 in peripheral blood of each group were detected by immunohistochemical method.Results:There was no significant difference in the levels of TNF-a and il-10 in the Xiaoshuan Decoction group and the physiological saline group at the first day after surgery(P>0.05).Compared with the Xiaoshuan Decoction group,the level of TNF-a in the Low molecular heparin sodium group decreased significantly,while the level of il-10 was significantly increased(P<0.05).Compared with the normal saline group,the level of TNF-a in the Xiaoshuan Decoction and the Low molecular heparin sodium group was significantly decreased,while the level of il-10 was significantly increased and its ratio decreased at the third day after surgery(P < 0.05),and there was no significant difference in the levels of TNF-a and il-10 in the low molecular heparin sodium group at the third and seventh day after surgery(P>0.05).Conclusion:The Xiaoshuan Decoction can effectively elevate the level of IL-10,and reduce the level of TNF-α,and thus reversing Th1/Th2 imbalance to achieve therapeutic effect.
