Research Background:Ischemic Stroke(IS)is a common type of stroke,it has caused more and more attention because of high incidence,high morbidity and other factors.For long time,most studies used a single target to protect the neuronal structure and prevent its apoptosis,but later it is gradually realized that the treatment of a single target is far from enough to protect he death of neurons.The microenvironment and the surrounding cells and the control of the surrounding cells,and a single target treatment can only affect one or several related factors,and it can’t fully control the neuronal damage after stroke pathways.At present,we propose a multi-target treatment plan for structural damage of Neurovascular Unit(NVU),which will have a better effect on the recovery and prognosis of post-stroke neurological function.Among them,the recovery of neurological function after cerebral ischemia mainly depends on the regeneration of synapses between neurons and the formation of new neural pathways in uncontrollable synapses,namely synaptic plasticity.The plasticity of synapses can be measured in two ways,the plasticity of the synaptic morphology and the plasticity of the function.According to the "field" hypothesis,the tufting method is widely used to regulate the structure of the brain.The method is based on the,field" hypothesis,which is based on the basic theory and the clinical practice.This study is based on the National Natural Science Foundation of China,which aims to observe the changes of neurological function,the plasticity of synaptic membrane in ischemic penumbra,the molecular mechanismObjective:To observe the effect of scalp acupuncture on neurological function score and the changes of synaptic ultrastructure in ischemic penumbra and the expression of GAP-43 in cerebral infarct area Methods:Mechanisms of synaptic plasticity in ischemic penumbra of stroke.Method:The research purchased 72 healthy male SD rates,250-280gand were divided into cluster scalp acupuncture group A,model group B and sham operation group C by random number table.And each group was divided into day 1、day 7 and day 14 three groups by treatment time.MCAO model was made in Group A and B by common carotid artery puncture method.Group C was only isolated the common carotid artery.Group A was treated by cluster scalp acupuncture everyday while group B and C didn’t do any treatment.These three groups were raised in a routine way.All rats assessed with neurological scores after 4 hours by model surgery and before draw materials time.The brain tissues were dehydrated,stained and sectioned.The density of postsynaptic density,the width of synaptic cleft and the curvature of synaptic interface were observed by transmission electron microscopy(TEM).Each time point,Six rats were sacrificed at each time point,saline was perfused,and the brain was stored in-80 ℃refrigerator.The content of GAP-43 protein in cerebral infarct area was detected by western-blot.Results:1.Neurological scoresOn 4 hours:group A and B compared with group C was statistically significant(P<0.05),which showed that the MCAO model was successfully made.Group A compared with group B was not statistically significant(P>0.05),which means the two groups were comparable.On day 1:the neurological score of group A was slightly better than that of group B,but there was no statistical significance(P>0.05).This indicates showed that although cluster scalp acupuncture treatment for 1 day has some positive effects,but there were no significant differences.On day 7 and day 14:group A and B were statistically significant(P<0.05),which means the cluster scalp acupuncture has a significant effect on the behavioral symptoms of MCAO rats,and has positive significance on the motor function recovery.2.Observation of synaptic ultrastructure and Gap-43 contentOn day 1:There was no significant difference in synapse thickness,synaptic interface curvature,synaptic gap width and GAP-43 in group A and B compared with group C(P>0.05).On day 7:the synaptic cleft width and synaptic interface curvature were significantly higher than those in group B(P<0.05).There was no significant difference in synaptic thickness and GAP-43 between group A and group B(P>0.05).Conclusion:Scalp acupuncture can improve the plasticity of synaptic structure and function in ischemic penumbra region of rats after MCAO.On day 14:Compared with group B,the thickness of synaptic density increased and the width of synaptic cleft was decreased Statistical differences(P>0.05).The content of GAP-43 in group A was higher than that in group B,but the result was not significant(P<0.05).There was no significant difference in the curvature of the synaptic interface between the three groups(P>0.05).Conclusion:1.Cluster scalp acupuncture can promote the recovery of neurological function in MCAO rats and improve the related behavioral indicators.2.Cluster scalp acupuncture can reduce the width of synaptic intervertebral space in the penumbra of MCAO rats,increase the thickness of postsynaptic densities,improve the synaptic plasticity after cerebral ischemia,and contribute
