Research Background:Because of its high incidence, high mortality and high disability rate, AIS has been paid more and more attention. In this regard, large numbers of research have been down, but for now the mechanism is still not very clear. NVU is a very popular research aspect in recent years. It emphasizes that as a whole, the interaction of neurons, vascular endothelial cells, astrocytes, microglia, matrix proteins, and so on, has an impact on the pathology and physiology of the disease. This study relies on the National Natural Science Foundation of China "The mechanism study on the effect of the neurovascular unit remodeling after cerebral ischemia by the cluster needling of scalp acupuncture", and mainly through the observation and statistical data of the changes of the neurological scores and the expression of new blood vessels in the ischemic penumbra area of MCAO rats treated by cluster scalp acupuncture, in order to explain the process and mechanism of the pathological changes.Objective:To observe the effect of cluster scalp acupuncture on the neurological scores of MCAO rats, and to observe the effect of cluster scalp acupuncture on the expression of CD34 microvessel density in the ischemic penumbra area of MCAO rats.Method:The research purchased 54 healthy male SD rates,250-280g and were divided into model group A, sham operation group B and cluster scalp acupuncture group C by random number table. And each group was divided into day 1, day 7 and day 14 three groups by treatment time. MCAO model was made in Group A and C by common carotid artery puncture method. Group B was only isolated the common carotid artery. Group C was treated by cluster scalp acupuncture everyday while group A and B didn’t do any treatment. These three groups were raised in a routine way. All rats assessed with neurological scores after 4 hours by model surgery and before draw materials time. The rat’s brains were collected after perfusion. These samples were paraffin embedded and made into sections. Immunofluorescence was used to dye CD34 antigen. The expression of CD34 in the ischemic penumbra area of MCAO rats was observed by fluorescence microscopy.Results:1. Neurological scoresOn 4 hours:group A and C compared with group B was statistically significant (P<0.05), which showed that the MCAO model was successfully made and the groups were successfully divided. Group A compared with group C was not statistically significant (P>0.05), which means the two groups were comparable.On day 1:the neurological score of group C was slightly better than that of group A, but there was no statistical significance (P>0.05). This indicates showed that although cluster scalp acupuncture treatment for 1 day has some positive effects, but there were no significant differences.On day 7 and day 14:group A and C were statistically significant (P<0.05), which means the cluster scalp acupuncture has a significant effect on the behavioral symptoms of MCAO rats, and has positive significance on the motor function recovery.2. Microvessel density:On day 1:the CD34 microvessel density is lower than group B compared with group A and C, but there was no statistical significance (P>0.05), probably because the ischemic penumbra area of ischemic necrosis of a large number of blood vessels, and the newly formed vascular endothelial cells have not yet formed vascular morphology. Group C has a slightly higher microvessel density than A group (P<0.05), which indicates that the early cluster scalp acupuncture treatment has protective effects on the blood vessels in the ischemic penumbra area.On day 7:group C was significantly higher than group A with statistical significance (P<0.05). It was showed that cluster scalp acupuncture can promote angiogenesis in the ischemic area of MCAO rats.On day 14:group C continues to rise, while group A was significantly reduced, and group C and A has statistical significance compared to group B (P<0.05). This means that cluster scalp acupuncture can extend new blood vessels, and promote the formation of new blood vessels.3. Through the fluorescence microscopeWe can see that the infarct area almost had no new blood vessels. New blood vessels were densely distributed in the ischemic penumbra area around. The vascular pattern is irregular, and to surround the infarct area.Conclusion:1. Cluster scalp acupuncture can improve the behavioral symptoms of MCAO rats and promote the recovery of limb motor function.2. Cluster scalp acupuncture can increase the microvascular density and promote the formation of new blood vessels in MCAO rats.
