Objective:To observe the effect of Carapax Trionycis-zedoary turmeric on β-catenin,c-Myc and cyclin D1 proteins of Wnt/ β-catenin signal pathway in MCF-7 human breast cancer cells,and to explore the inhibitory effect of Carapax Trionyciszedoary turmeric on the proliferation of MCF-7 human breast cancer cells and its possible mechanism.Method:55 SPF female SD rats weighing 240-260 g were randomly divided into 4 groups:Carapax Trionycis group(n = 10),zedoary turmeric group(n = 10),Carapax Trionycis-zedoary turmeric medicine group(n =10)and control group(n = 25).After 7 days,the rats were killed and the serum was kept for reserve.The stored serum and the DMEM medium are combined into different doses of drug-containing serum medium with a volume fraction of 10%,and the MCF-7 cells are treated.They were marked as: control serum group,Carapax Trionycis low dose group,Carapax Trionycis medium dose group,Carapax Trionycis high dose group,Rhizoma Zedoariae low dose group,Zedoary turmeric high dose group,Carapax Trionycis zedoary medicine pair low dose group,Carapax Trionycis zedoary medicine pair middle dose group,Carapax Trionyciszedoary medicine pair high dose group,positive control group.After incubated in the incubator for 48 h and 72 h,the inhibition rate of cell proliferation was detected by MTT assay.After incubated in the incubator for 48 hours,the cell cycle was detected by flow cytometry and the expression of β-Catenin,c-Myc and cyclin D1 protein was detected by Western blot.Result: 1.Compared with the control serum group,each drug-containing serum group could inhibit the proliferation of MCF-7 cells after 48 h and 72 h,and the inhibition rate was as follows: high-dose group > middle-dose group > low-dose group(P < 0.05),and the inhibition rate of each high-dose group was as follows: Carapax Trionycis-zedoary turmeric drug on high-dose group > zedoary turmeric high-dose group > Carapax Trionycis high-dose group(P < 0.05).2.Compared with the control serum group,the proportion of G0/G1 phase cells in each drug-containing serum group increased significantly after 48 hours(P < 0.05),and the proportion of G0 / G1 phase cells in each drug-containing serum group was as follows: high-dose group > middle-dose group > low-dose group(P < 0.05).The proportion of S phase cells in high-dose group < medium-dose group < low-dose group(P < 0.05),and the proportion of G0 / G1 phase cells in each drug-containing serum group was as follows: high-dose group > mediumdose group > low-dose group(P < 0.05).The proportion of G0 / G1 phase cells in each high dose group was as follows: Carapax Trionycis-zedoary turmeric group > zedoary turmeric high-dose group > Carapax Trionycis high-dose group(P < 0.05).The proportion of S phase cells in Carapax Trionycis-zedoary turmeric group was: Carapax Trionycis-zedoary turmeric high-dose group < zedoary turmeric high-dose group <Carapax Trionycis high-dose group(P < 0.05).3.Compared with the control serum group,the protein expression of β-Catenin,c-Myc and cyclin D1 decreased.The protein expression in each drug-containing serum group was as follows: high dose group < middle dose group < low dose group(P < 0.05),and the protein expression of each high dose group was as follows:Carapax Trionycis-zedoary turmeric group < zedoary turmeric high dose group < Carapax Trionycis high dose group.Conclusion:1.The drug-containing serum of all groups had inhibitory effect on the proliferation of MCF-7 cells,and the inhibitory effect was the best in the high-dose group compared with different doses of the same traditional Chinese medicine,The inhibitory effect of Carapax Trionycis-zedoary turmeric was better than that of single drug.2.The drug-containing serum,the antidote serum,the amoeba-serotonin to the drug-containing serum effect MCF-7 cells can prevent g1/G0 stage of the cell smoothly into the S and G2 stage,so that the MCF-7 cell fissure process is disturbed,resulting in its proliferation inhibited;3.The mechanism of anti-MCF-7 effect of Carapax Trionycis-zedoary turmeric on human breast cancer cells may be related to the inhibition of the expression of β-catenin in Wnt/ β-catenin signal pathway and the down-regulation of β-catenin and the downstream target gene Cyclin D1,c-Myc protein expression of β-catenin.
