Objective:To investigate the relationship between the expression of related proteins p-PI3 K and p-AKT in PI3 K / AKT signal transduction pathway in gastric cancer cells and related pathological parameters of gastric cancer,so as to provide a new idea for the diagnosis and treatment of gastric cancer.Methods:A total of 100 gastric cancer patients were collected from the Department of Gastrointestinal surgery of the second affiliated Hospital of Nanchang University,including 61 males and 39 females,aged from 35 to 78 years old,with an average age of 63.According to the eighth edition of AJCC TNM staging criteria for gastric cancer,there were 9 cases of stage I,33 cases of II stage,51 cases of III stage and 7cases of IV stage.The degree of differentiation: 41 cases of low differentiation,46 cases of middle differentiation,13 cases of high differentiation;depth of invasion: 63 cases of lesions penetrated serosa,37 cases did not;lymph node metastasis: 72 cases of gastric cancer had lymph node metastasis,28 cases had no lymph node metastasis;at the same time,a total of 100 cases of paracancerous tissues were taken(5cm from the cutting edge of the tumor).The gastric mucosal tissues of 48 patients with normal gastroscopy were taken.Among the 48 examiners,there were 26 males and 22 females,aged from 31 to 68 years old,with an average age of 58 years.The expression of p-PI3 K and p-AKT in gastric cancer,paracancerous tissue and normal gastric mucosa was detected by immunofluorescence.The related data were collected and analyzed.By comparing the expression of two related proteins in three groups of different cells,the relationship between the expression of PI3K/Akt signaling pathway related proteins and gastric cancer related pathological parameters was discussed.So as to further clarify the role of PI3K/Akt signal pathway in the occurrence and development of gastric cancer.Results:The expression rate of p-PI3 K in gastric cancer cells was 55.0%,the expression rates in paracancerous tissues and normal gastric mucosa cells were 23.0% and 14.6%respectively,and the expression rates of p-AKT in gastric cancer cells were 29.0%.The expression rates in paracancerous and normal gastric mucosa were 13.0% and8.33% respectively(P < 0.05).The two related proteins showed a significant downward trend in gastric cancer tissue cells,paracancerous tissue cells and normal gastric mucosal cells,and there were significant differences among the three tissue cells(P < 0.05).By analyzing the expression differences of the two proteins in different gastric cancer cells under different pathological parameters,we found that there was no significant correlation between the expression of p-PI3 K and p-AKT in gastric cancer tissues and the sex and age of the patients.However,it was significantly correlated with the TNM stage,the degree of differentiation,the depth of invasion and the presence of lymph node metastasis(P < 0.05).Conclusion:Compared with paracancerous tissues and normal gastric mucosa tissues,PI3K/AKT-related proteins p-PI3 K and p-AKT are highly expressed in gastric cancer tissues.The expression of PI3 K / AKT-related proteins p-PI3 K and p-AKT in gastric cancer cells had no significant correlation with age and sex,but had significant correlation with TNM stage,degree of differentiation and depth of invasion(p >0.05).The abnormal activation of PI3K/AKT signal transduction pathway may be a factor in the carcinogenesis of gastric mucosal cells,and the activated PI3K/AKT signal may accelerate the development of gastric cancer.The detection of p-PI3 K and p-AKT may play a certain role in screening,predicting the biological behavior and prognosis of early gastric cancer.At the same time,the development of targeted drugs for PI3K/AKT signal pathway can be used as a new idea for the treatment of gastric cancer.
