Objective:Chinese medicine treatment because of its treatment with Western medicine together to enhance the anti-tumor effect,and to reduce the impact of drugs on normal human life activities,especially for postoperative and radiotherapy and chemotherapy rehabilitation treatment,advanced gastric cancer patients support Treatment,have a better effect.Professor Ge Xinguo of Nanjing University of Traditional Chinese Medicine in the treatment of gastric cancer prescription Gastric cancer I decoction,in the clinical application process,has a good control of tumor growth,reducing postoperative recurrence and metastasis rate,extending PFS,but its mechanism is not clear.To study the effect of in vitro on gastric cancer cell line SGC-7901 in vitro and its effect on intracellular PI3K/AKT signaling pathway and its effect on intracellular PTEN secretion.Exploring Gastric cancer I decoction’s mechanism of action can provide new ideas and methods for the comprehensive treatment of gastric cancer and benefit the majority of patients with gastric cancer.Methods:Gastric cancer cells SGC-7901 were cultured in vitro and the drug containing serum of Gastric cancer I decoction was made in New Zealand rabbits.Set different concentrations of drug-containing serum to act on gastric cancer cells SGC-7901.The CCK8 method was used to analyze the effect of drug containing serum on the proliferation of gastric cancer cells SGC-7901.The effect of drug-containing serum on cell migration was determined by scratch test.The effect of drug-containing serum on the cell cycle was measured by flow cytometer.Western blot was used to determine the effect of drug-containing serum on the expression of PTEN,PI3K and p-AKT proteins.Results:Gastric cancer I decoction could inhibit the proliferation of gastric cancer cells SGC-7901(p<0.01)and inhibit the effect of migration(p<0.05)in a dose-and time-dependent manner.When 10%concentration of drug-containing serum affected gastric cancer cell SGC-7901,the gastric cells blocked in the GO/G1 phase,hinder the cell mitosis.Western blot analysis showed that the expression of p-AKT and P13K was inhibited in the experimental group,while the expression rate of PTEN protein increased(p<0.01).Conclusion:1.Gastric cancer I decoction could inhibit the proliferation and migration of SGC-7901 cells in vitro.2.Gastric cancer I decoction can block the cell cycle of SGC-7901 cells in GO/G1 phase.3.Gastric cancer I decoction can increase the secretion of PTEN protein and down-regulate the PI3K/AKT signaling pathway to play an anti-tumor effect.
