| Objective:The purpose of this study was to detect the differences in PLCE1 protein expression in Xinjiang oral squamous cell carcinoma tissues and adjacent normal tissues by immunohistochemical Envision method,and to analyze the expression of PLCE1 protein and the clinicopathological characteristics of oral squamous cell carcinoma patients and their prognosis.It provides a theoretical basis and experimental basis for the early diagnosis and treatment of oral squamous cell carcinoma and the better judgment of the prognosis of patients with oral squamous cell carcinoma.Methods:From 2004 to 2018,124 cases of pathologically confirmed oral squamous cell carcinoma tissues and 63 normal tissue specimens adjacent to the cancer were collected from the Department of Pathology of the First Affiliated Hospital of Shihezi University Medical College,Xinjiang,and clinical pathological and prognostic data were collected.Immunohistochemical Envision method was used to detect the expression of PLCE1 protein in oral squamous cell carcinoma tissues after preparing tissue chips from each tissue specimen.Statistics and analysis of PLCE1 expression in oral squamous cell carcinoma tissues and its correlation with oral squamous cells Correlation between clinicopathological data and prognosis of cancer patients.Results:(1)PLCE1 was mainly located in the cytoplasm.The high expression rates of PLCE1protein in OSCC tissues and adjacent tissues were 41.9%(52/124)and 0%(0/63).(?~2=36.596,P<0.0001).There was no significant correlation between PLCE1 positive expres-sion and age,gender,tumor differentiation,T stage,lymph node metastasis and clinical stage of OSCC patients(P>0.05).(2)The area under the ROC curve(AUC)of PLCE1 expression in the diagnosis of oral squamous cell carcinoma was 0.954(95%CI,0.925-0.982),the optimal cut-off value was 7,and the sensitivity and specificity were 83.9%and 95.2%,respectively.(3)The results of Kaplan-Meier analysis showed that the survival time of patients with high expression of PLCE1 was significantly different from that of patients with low expression of PLCE1(?~2=8.416,P=0.004).(4)Cox univariate analysis showed that PLCE1expression(P=0.006)and T stage(P=0.004)were the risk factors affecting the prognosis of patients;Cox multivariate analysis results further showed PLCE1 expression(P=0.008)and T stage(P=0.005)is an independent factor for the prognosis of patients.Conclusion:PLCE1 protein is highly expressed in Xinjiang oral squamous cell carcinoma tissues and it is associated with poor prognosis of patients.It is expected to become a new biomarker for OSCC diagnosis and prognosis prediction. |
