Objective:To study the effects of lycium barbarum polysaccharide(LBP)on the growth,proliferation and biological activity of schwann cells(SCs)required for repair of peripheral nerve injury.To study the effect of LBP combined with acellular nerve xenograft(ANX)in repairing sciatic nerve defects in rats.Methods: To obtain the SCs of the sciatic nerve of SD rats,and used the differential adherence method to cultivate the SCs of SD rats.Divide lycium barbarum polysaccharides into seven groups: 0ug / ml,50ug / ml,100ug / ml,150ug / ml,200ug / ml,250ug / ml,500 ug / ml,and mix with schwann cells respectively.On days 1,3,5,7,and 9 after Schwann cells were cultured in Petri dishes,the biological activity of SCs in each group was detected by CCK-8 method,and the gene expression levels of nerve growth factor(NGF),brainderived neurotrophic factor(BDNF)and basic fibroblast growth factor(b FGF)were detected by rt-PCR.To screen the optimal LBP concentration of cultivating SCs.The SD rats were divided into three groups(n = 10): ANX group,ANX-SCs group and ANX-LBP-SCs group.A rat model of sciatic nerve defects was prepared under sterile conditions.The sciatic nerve defect of each rat is about 10mm.Select the tibial nerve of the rabbit to make ANX.Eight weeks after surgery,hot plate experiments,sciatic nerve function index(SFI)and motor nerve conduction velocity(MCV)of the affected limbs in each group were detected.In addition,the nerve fibers and myelin sheath regeneration in nerve grafts of three groups were compared by projection electron microscopy.Results: In vitro experiment,the concentration of LBP tending to 150-250 ug / ml was statistically significant compared with other concentrations(P <0.05),and the activity of the remaining groups of cells was not statistically different between the two groups(P> 0.05).On Days 2-4,there was no significant difference in the activities of the seven groups(P>0.05).The m RNA expression of BDNF,NGF and b FGF in the 200ug / ml LBP-SCs group was detected by rt-PCR,which was statistically significant compared with other groups(P<0.05).Eight weeks after transplantation,the ANX-LBP-SCs group was found to have better hot plate experiments,SFI detection,and nerve conduction velocity.Transmission electron microscopy showed that myelination and nerve fiber regeneration were significant in the ANX-LBP-SCs group.Conclusions: When the concentration of LBP is 200ug / ml,it can promote the proliferation of SCs and secrete a large amount of neurotrophic factors;2.SCs culture system containing LBP combined with ANX transplantation can significantly promote peripheral nerve regeneration.
