Selenizing polysaccharide has many functions such as anti-cancer,anti-oxidation,lowering blood sugar,enhancing immunity and so on,and its biological activity is higher than that of polysaccharide and selenium which act alone.In this experiment,lycium barbarum polysaccharide(LBP)was selenized by nitric acid-sodium selenite method,and the antioxidative activity and hepatoprotective effect of the LBP and selenizing LBP were compared.The aim is to improve the bioavailability of LBP and provide a theoretical basis for the development of new anti-oxidation and hepatoprotective agents.The results show:1.Two kinds of selenizing LBP were sLBPp and sLBPpl,their yield were 32.43%and 30.05%,carbohydrate content were 51.63%and 54.91%,selenium content were 11.16 mg/g and 10.75 mg/g,respectively.And the infrared spectra of two kinds of selenizing polysaccharide demonstrated the bond of selenite selenium ester and selenium ester.2.The scavenging ability of DPPH free radicals,hydroxyl free radicals and ABTS free radicals of sLBPp and sLBPpl were significantly higher than those of LBP control group in vitro(P<0.05);The GSH-Px activity and SOD activity in serum were significantly higher than those of LBP control group(P<0.05),and the MDA content was significantly lower than that of LBP control group(P<0.05)in vivo.3.The sLBPp could significantly reduce the content of ALT,AST and ALP in serum of mice with liver injury induced by CCl4(P<0.05);and significantly reduce the content of MDA,improve the activity of SOD,GSH-Px and T-AOC in liver homogenate(P<0.05),the effects were stronger than those of LBP.Conclusion:1.The sLBPp was successfully modified by nitric acid-sodium selenite method,the reaction conditions were 400 mg of sodium selenite for 500 mg of LBPp,LBPp1,reaction temperature of 70℃ and reaction time of 6 h.2.Selenization modification can significantly improve the antioxidant activity of LBP in vitro(DPPH free radicals,hydroxyl radicals and ABTS free radical scavenging ability),in vivo(increased GSH-Px and SOD activities,and decreased MDA content).3.sLBPp acted a significantly hepatoprotective effect by reducing the contents of ALT,AST,ALP and MDA and increasing the activity of TP,SOD,GSH-Px and T-AOC.
