| Objective:Through the establishment of autism drug network and the establishment of autism"drug-target" network,through network analysis and computer technology,provide potential targets and drugs for the future treatment of autism,and recommend reasonable new therapy for autism patients.Data and methods:In this paper,taking the drugbank whole drug database as the data source,we extract 2527 drug data and 3786 target data through python.Based on 2527 drug data,the drug network was established,the network characteristics were analyzed,and the drug network was cluster analyzed.By observing the drug classification characteristics in each drug community,some drugs with obvious differences from the drug indications in the community were found(except for clinical drugs with autism).Then,by consulting the relevant materials,we can find out whether they are related to the community owners In order to have correlation between indications,drugs related to autism treatment are selected as potential treatment drugs through autism related pathogenesis.Based on 2527 drug data and 3786 target data,a "drug target" binary network is established,and a binary network recommendation algorithm based on material diffusion is selected to carry out autism related drug and target recommendation,so as to provide ideas for discovering the potential value of autism treatment.Results:By analyzing the characteristics of drug network and clustering analysis,we found that five drugs showed good drug value attributes,which were zinc(zinc),amitriptyline(amitriptyline),olanzapine(olanzapine),clozapine(clozapine)and imipramine(cimetico).These drugs are widely used on a good basis,and can play a bridge or transitional role in the combination of drugs.Zinc(zinc)can be used in the treatment of children’s growth retardation,which is an important clinical drug for autism.In the process of target and drug recommendation for the "drug target" binary network,8 of the 21 recommended targets for 7 important autistic drugs are d(1a)dopamine receptor,D(2)dopamine receptor,D(3)dopamine receptor,D(4)dopamine receptor,alpha-Id active receptor,alpha-2a active receptor,5-hydroxytryptamine receptor 1A.Peroxisome proportioner activated receptor gamma coincides with the existing targets of important autistic drugs.Through drug recommendation for autism related targets,10 of the recommended drugs coincide with clinical drugs for autism,which are:chlorpromazine,clozapine,olanzapine,imipramine,aripiprazole,quetiapine,galantamine,amitriptyline,vitamin C,zinc.Aripiprazole coincides with important autistic drugs.Based on the above,the results of drug and target recommendation have reference value.Conclusion:Based on the drug network,28 potential autistic drugs were found by social network analysis and cluster analysis.Based on the "drug target" association network,21 potential autistic treatment targets are recommended for 7 important autistic treatment drugs and 215 potential autistic treatment drugs are recommended for 73 autistic treatment drugs through the binary network recommendation algorithm.The potential therapeutic drugs for autism discovered through the drug network and the potential therapeutic targets and drugs recommended through the "drug target"associated network can provide ideas for discovering the potential value of the field of autism treatment and provide reference for the diagnosis and treatment of diseases,precision medicine and other related research. |
