Acute renal Injury(AKI)is characterized with acute renal dysfunction and renal tubular epithelial cell Injury,however,Ischemia Reperfusion Injury is a major cause of AKI.This study based on the unilateral renal ischemia reperfusion model in ICR male mice proved that the cellular stress response represented by oxidative stress and endoplasmic reticulum stress was extensively involved in ischemia/reperfusion induced AKI in mice by detecting the changes of oxidative stress and endoplasmic reticulum stress related indicators in kidney tissue and serum of mice in the control group and the cholecalciferol pretreatment group.Moreover,cholecalciferol pretreatment can significantly down-regulate oxidative stress and endoplasmic reticulum stress in renal tubular epithelial cells during renal ischemia reperfusion injury in mice,and finally effectively alleviate AKI induced by ischemia reperfusion injury.In this study,animal experiments and molecular biology experiments were used to prove that pretreatment with cholecalciferol can effectively alleviate renal pathological damage and impaired renal filtration function caused by renal ischemia reperfusion injury in ICR male mice.And by detecting the changes of oxidative stress related genes and protein expression levels,it was proved that pretreatment with cholecalciferol can reduce the degree of oxidative stress during renal ischemia reperfusion in ICR male mice.Moreover,this study further demonstrated that the pretreatment with cholecalciferol can effectively relieve the stress degree of endoplasmic reticulum during renal ischemia reperfusion in ICR male mice.Furthermore,the expression level of related indexes in the downstream stress pathway of endoplasmic reticulum was detected.On the other hand,this study also demonstrated that pretreatment with cholecalciferol can effectively inhibit the apoptosis of renal cells induced by renal ischemia reperfusion injury in mice.In summary,this study aims to demonstrate the protective effect of cholecalciferol pretreatment on acute renal injury induced by renal ischemia reperfusion in mice.Furthermore,the down-regulation effects and mechanisms of cholecalciferol pretreatment on cellular stress response in renal cells of mice were further demonstrated.
