| Objective: To investigate the expression and correlation of ciRS-7,miR-7 and NF-κB in GC tissues.Methods: qRT-PCR technique was used to detect the expression of ciRS-7,miR-7,and the expression levels of NF-κB p65 protein were detected by immunohistochemistry in 20 cases of GC and its para-carcinoma tissues.Analyze the relationship between the three and the clinicopathological characteristics of patients.Results:(1)The expression of ciRS-7 in GC tissues was significantly higher than that in matched para-carcinoma tissues(P < 0.05),and its expression level correlated with the lymph node metastasis(P=0.035)and clinical stage(P=0.028),but not with age,sex,tumor diameter and the invasive depth of GC patients(P > 0.05).(2)The expression of miR-7 in GC tissues was down-regulated than that in matched para-carcinoma tissues(P < 0.05),and its expression level correlated with the invasive depth(P=0.024),lymph node metastasis(P=0.024)and clinical stage(P=0.008),but not with age,sex and tumor diameter of GC patients(P > 0.05).(3)The expression level of NF-κB p65 protein in GC tissues was significantly higher than that in para-carcinoma tissues(P < 0.05),and correlated with the invasive depth(P=0.024),lymph node metastasis(P=0.007)and clinical stage(P=0.008),but not related with age,gender and tumor size of GC patients(P > 0.05).(4)The expression leveal of cirs-7 was negatively correlated with mir-7 in GC tissues(r =-0.245),but there was no statistical difference(P > 0.05).In GC tissues,the expression of NF-κB p65 protein was positively correlated with the expression of ciRS-7(r= 0.692),but negatively correlated with the expression of miR-7(r =-0.478),the difference was statistically significant(P < 0.05).Conclusions:(1)Compared with the adjacent tissues,the expression of ciRS-7 and NF-κB are up-regulated in GC tissues,while the expression of miR-7 is down-regulated.(2)In GC tissues,the expression of ciRS-7 is negatively correlated with that of miR-7 to some extent.The expression of NF-κB p65 is positively correlated with the expression of ciRS-7,and negatively correlated with the expression level of miR-7.(3)In GC tissues,the expression levels of ciRS-7 is closely related to lymph node metastasis,and clinical stageof GC patients,while the expression levels of miR-7 and NF-κB are closely related to invasive depth,lymph node metastasis,and clinical stage of GC patients.It is helpful to evaluate the infiltration,metastasis and prognosis of GC patients through combined detection of ciRS-7,miR-7 and NF-κB. |
